以天津市某花圃栽培中发现的一种新型病害为研究对象,采用形态特征观察、致病性测定及rDNA-ITS测序、比对研究方法,对其进行了病原鉴定,并对病原菌的生物学特性进行了简单研究。结果表明:该菌株在PDA培养基上4d菌落直径达到34.6~37.2mm,具明显的灰白色轮纹。分生孢子呈梭形,宽度为4.83~9.11μm,由5个细胞组成,细胞间有隔膜,孢子顶端有2~3根附属丝,长度约为5.93~36.23μm,尾胞具附属丝1根,长度为2.85~16.05μm。其rDNA-ITS测序比对结果表明,该菌株与棒状拟盘多毛孢菌Pestalotiopsisclavispora的同源性达到99%。基于形态学特征和rDNA-ITS测序比对结果,该病原菌被最终鉴定为Pestalotiopsis clavispora(Arth.)Stey.,经检索月季病害,属国内首次发现由棒状拟盘多毛孢菌引起月季叶枯病。生物学特性研究表明,该菌生长最适温度是25℃,但20~30℃范围内差异不显著;适宜pH5.0;最佳碳源为乳糖。
Taking a new type of plant disease found in a flower garden in Tianjin as the research object, morphological observation, pathogenicity determination, rDNA-ITS sequencing and comparison methods were carried out to identify the pathogen, and the pathogenic bacteria biology Characteristics of a simple study. Based on the morphological characteristics and sequencing results of rDNA-ITS, the pathogen was finally identified as Pestalotiopsis clavispora (Arth.) Stey. After searching for the disease of rose, it was the first discovery in China that it caused the leaf blight caused by P. crenulatum. The study of biological characteristics showed that the optimal temperature for growth of the strain was 25 ℃, but the difference was not significant in the range of 20-30 ℃; the optimum pH was 5.0; and the optimum carbon source was lactose.
