目的:观察安肠愈疡汤口服合加味生肌散灌肠对实验性溃疡性结肠炎(UC)大鼠的疗效及机制探讨。方法:将90只SD大鼠随机分为6组[正常对照组、模型对照组、安肠愈疡汤组、加味生肌散组、安肠愈疡汤合加味生肌散组、柳氮磺吡啶(SASP)组]。除正常对照组外,其他各组均采用2,4,6-三硝酸苯磺酸复制UC模型,给药3周后,进行结肠大体形态和组织损伤评分,测定结肠组织内髓过氧化物酶(MPO)的活性以及白介素-1β(IL-1β)、白介素-8(IL-8)、肿瘤坏死因子(TNF-α)、白介素-10(IL-10)的含量。结果:各试验组大鼠结肠黏膜均有不同程度的炎症修复、溃疡愈合。结肠大体形态和组织学损伤评分及MPO活性均不同程度降低,结肠黏膜中IL-1β、IL-8、TNF-α含量也不同程度降低,而IL-10含量升高,与模型对照组相比有显著性差异(P<0.05或P<0.01),尤其以安肠愈疡汤合加味生肌散组最显著。结论:安肠愈疡汤合加味生肌散具有较好的抗炎和促进溃疡修复的作用,并能下调致炎细胞因子和上调抗炎细胞因子,从而使免疫功能恢复。
Objective: To observe the efficacy and mechanism of Anchang Yuyang Decoction combined with Jiawei Shengjisan enema on experimental ulcerative colitis (UC) rats. Methods: 90 SD rats were randomly divided into 6 groups: normal control group, model control group, Anju Yuyang Decoction group, Jiawei Shengjisan group, Anju Yuyang Decoction, Jiawei Shengjisan group, and sulfasalazine. Pyridine (SASP) group]. In addition to the normal control group, the other groups were replicated with 2,4,6-trinitrobenzenesulfonic acid UC model. After 3 weeks of administration, gross morphology and tissue damage scores of the colon were scored to determine the myeloperoxidase in the colon tissue. (MPO) activity and levels of interleukin-1β (IL-1β), interleukin-8 (IL-8), tumor necrosis factor (TNF-α), and interleukin-10 (IL-10). RESULTS: All rats in the experimental group had different degree of inflammatory repair and ulcer healing. The gross morphology, histological damage scores and MPO activity of the colon were reduced to varying degrees. The contents of IL-1β, IL-8, and TNF-α in the colonic mucosa were also decreased to varying degrees, while the IL-10 levels were increased, compared with the model control group. There was a significant difference (P<0.05 or P<0.01), especially in the Anjie Yuyang Decoction plus flavored Shengjisan group. Conclusion: Anjie Yuyang Decoction plus Modified Shengjisan has a good anti-inflammatory and ulcer-promoting effect, and can down-regulate pro-inflammatory cytokines and up-regulate anti-inflammatory cytokines, so that the immune function can be restored.
