目的 探讨胎盘绒毛膜在胎儿期造血中的作用 ,寻找造血干 /祖细胞的新来源 ,供临床移植应用。方法 分别测定并比较脐动、静脉血中CD+34 细胞含量及粒 单细胞集落形成单位(CFU GM)、混合集落形成单位 (CFU MIX)、高增殖潜能集落形成单位 (HPP CFU)集落的产率。结果脐静脉中CD+34 及CD+34 /CD-38细胞含量分别为 (1 2 5± 0 94) %和 (0 6 1± 0 5 0 ) %,较脐动脉血中的含量(0 77± 0 5 0 ) %、(0 33± 0 2 7) %显著增高 (P均 <0 0 5 ) ;脐动、静脉血单个核细胞 (MNC)大多处于静止期 (>95 %) ,二者差异无显著性 (P >0 0 5 ) ,但合成期细胞的比率在脐静脉血中为 (2 2± 1 8) %,明显高于脐动脉血的比率 (0 9± 1 0 ) %;脐静脉血中CFU GM、CFU MIX和HPP CFU的产率分别为 (4 9 7± 2 8) / 2× 10 5MNC、(8 7± 4 6 ) / 1× 10 5MNC和 (1 6± 1 2 ) / 1× 10 5MNC ,明显高于脐动脉血的产率 (18±9) / 2× 10 5MNC、(5 2± 2 7) / 1× 10 5MNC和 (0 4± 0 4) / 1× 10 5MNC ,差异有显著性 (P均 <0 0 5 )。结论脐静脉血中造血干 /祖细胞的含量高于脐动脉 ,提示胎盘绒毛膜在胎儿期造血中具有一定的作用 ,并有可能成为造血干 /祖细胞的又一来源
Objective To investigate the role of placental chorion in the hematopoiesis of fetus and to search for a new source of hematopoietic stem / progenitor cells for clinical transplantation. Methods The levels of CD34 + cells and CFU GM, CFU MIX and HPP CFU colonies in umbilical artery and venous blood were measured and compared respectively rate. Results The levels of CD34 + and CD34 + CD38 in umbilical vein were (1 2 5 ± 0 94)% and (0 6 1 ± 0 5 0)%, respectively, which were significantly higher than those in umbilical artery (0 35 ± 0)% and (0 33 ± 0 2 7)%, respectively (all P 0 05). Most of the umbilical arterial and venous blood mononuclear cells (MNC) were in quiescence (> 95% There was no significant difference between the two groups (P> 0.05). However, the ratio of cells in the synthesis phase was (22 ± 18)% in umbilical blood and significantly higher than that in umbilical artery (0 9 ± 10%). The yields of CFU GM, CFU MIX and HPP CFU in umbilical cord blood were (497 ± 2 8) / 2 × 10 5 MNC, (8 7 ± 4 6) / 1 × 10 5 MNC and (16 ± 1 2) ) / 1 × 10 5MNC was significantly higher than that of umbilical artery (18 ± 9) / 2 × 10 5MNC, (5 2 ± 2 7) / 1 × 10 5MNC and (4 ± 0 4) / 1 × 10 5MNC, the difference was significant (P <0 05). Conclusion The content of hematopoietic stem / progenitor cells in umbilical vein blood is higher than that in umbilical artery, suggesting that placental chorion has a certain role in hematopoiesis during fetal period and may become another source of hematopoietic stem / progenitor cells
