目的:研究石榴皮鞣质对被动型Heymann肾炎大鼠的改善作用。方法:兔皮内多次免疫制备兔抗鼠Fx1A抗血清,然后尾静脉注射Fx1A抗血清,构建大鼠被动型Heymann肾炎(PHN)模型。将实验大鼠随机分为6组,为正常对照组、模型组、贝那普利组、石榴皮鞣质高、中以及低剂量组。造模成功后,每天用相应的药物进行灌胃给药治疗,持续5周,收集各组大鼠每周的尿液用于检测24 h尿蛋白。给药治疗结束后,收集各组大鼠的血清用于检测血清中的白蛋白、尿素氮、肌酐、总胆固醇和三酰甘油等指标;取肾脏,进行HE和Masson染色,观察石榴皮鞣质对PHN大鼠病理损伤的影响。结果:连续灌胃5周后,石榴皮鞣质可以明显降低被动型Heymann肾炎大鼠24 h尿蛋白含量(P<0.01),显著降低PHN大鼠血清肌酐(P<0.01)、尿素氮(P<0.01或P<0.05)、总胆固醇(P<0.01)以及三酰甘油(P<0.01或P<0.05)水平,并显著增高PHN模型大鼠血清白蛋白(P<0.01或P<0.05);HE和Masson染色结果表明,石榴皮鞣质对PHN模型大鼠病理损伤具有改善作用。结论:石榴皮鞣质对被动型Heymann肾炎大鼠具有改善作用。
Objective: To study the effect of pomegranate tannin on passive Heymann nephritis rats. METHODS: Rabbit anti-Fx1A antiserum was prepared by multiple immunization in rabbits. Fx1A antiserum was injected into tail vein to establish rat passive Heymann’s nephritis (PHN) model. The experimental rats were randomly divided into 6 groups, which were normal control group, model group, benazepril group, high, medium and low dose of pomegranate tannin. After the success of modeling, each day with the appropriate drug for gavage treatment for 5 weeks, each group of rats were collected weekly urine for 24 h urinary protein. After the treatment, the serum of rats in each group was collected for the detection of serum albumin, urea nitrogen, creatinine, total cholesterol and triglyceride. The kidneys were taken for HE and Masson staining, and the tannin On PHN rat pathological damage. Results: Serum creatinine (P <0.01), serum creatinine (P <0.01) and serum urea nitrogen (P <0.01) in PHN rats were decreased significantly (P <0.01 or P <0.05), total cholesterol (P <0.01) and triglyceride (P <0.01 or P <0.05), and significantly increased serum albumin (P <0.01 or P <0.05) HE and Masson staining results show that pomegranate tannin has an effect on pathological injury of PHN model rats. Conclusion: The tannin from pomegranate peel can improve the passive Heymann’s nephritis in rats.
