探究山梨猕猴桃Actinidia rufa根的化学成分。采用葡聚糖凝胶色谱法、薄层色谱硅胶色谱法、高效液相色谱法等方法分离得到9个化合物,鉴定为:2α,3β,19α,23,24-五羟基乌苏-12-烯-28-O-β-D-吡喃葡萄糖苷(1)、2α,3α,19α,24-四羟基乌苏-12-烯-28-O-β-D-吡喃葡萄糖苷(2)、2α,3α,24-三羟基乌苏-12-烯-28-酸(3)、2α,3α,24-三羟基齐墩果-12-烯-28-酸(4)、2α,3α,23,24-四羟基乌苏-12-烯-28-酸(5)、2α,3β,23,24-四羟基乌苏-12-烯-28-酸(6)、2α,3β,23-三羟基乌苏-12-烯-28-酸(7)、2α,3β,23-三羟基乌苏-12,20(30)-二烯-28-酸(8)、2α,3α,24-三羟基乌苏-12,20(30)-二烯-28-酸(9),其中化合物1和2为首次从该属植物中分离得到。细胞毒活性测试结果显示化合物2~4对人卵巢癌细胞株SKVO3和人甲状腺乳头状癌细胞株TPC-1具有强的细胞毒活性(IC50在10.99~16.41μmol·L~(-1)),而化合物3,4对人宫颈癌细胞株He La具有强的细胞毒活性,IC50分别为15.53,13.07μmol·L~(-1)。
To explore the chemical constituents of Actinidia rufa root in Yamanashi. Nine compounds were isolated by dextrane gel chromatography, thin-layer chromatography, silica gel chromatography and high performance liquid chromatography. The compounds were identified as 2α, 3β, 19α, 23,24-pentahydroxyunsu-12-ene -28-O-β-D-glucopyranoside (1), 2α, 3α, 19α, 24- tetrahydroxysussu-12-ene-28- 2α, 3α, 24-trihydroxyolean-12-en-28-oic acid (4), 2α, 3α, 23 (5), 2α, 3β, 23,24-tetrahydroxysusu-12-en-28-oic acid (6), 2α, 3β, 23-tris (8), 2α, 3α, 24-tris (23) -thiourea-12,20 (30) HydroxylUssu-12,20 (30) -diene-28-acid (9), in which compounds 1 and 2 were isolated from this genus for the first time. Cytotoxicity assay showed that compounds 2 ~ 4 have strong cytotoxic activity (IC50 of 10.99 ~ 16.41μmol·L -1) on human ovarian cancer cell line SKVO3 and human thyroid papillary carcinoma cell line TPC-1, Compound 3 and 4 showed strong cytotoxic activity against Hela cell lines with IC50 of 15.53 and 13.07 μmol·L -1, respectively.
