目的:考察N-三甲基壳聚糖对尼莫地平凝胶透皮吸收促进作用。方法:以季胺化程度为60%的N-三甲基壳聚糖(TMC60)为透皮吸收促进剂,以泊洛沙姆407为凝胶基质,制备尼莫地平凝胶(TMC60组)。用Franz扩散池进行体外透皮研究,并与合同浓度的氮酮(氮酮组)、油酸(油酸组)的尼莫地平凝胶及不含促进剂的尼莫地平凝胶(空白组)的效果进行比较。结果:空白组、氮酮组、TMC60组及油酸组的稳态透皮速率(J,μg·cm~(-2)·h~(-1))分别为6.59,13.22,16.39和16.77。与空白组相比,TMC60尼莫地平凝胶有显著的透皮吸收促进作用(P<0.05),且作用强度明显优于氮酮(P<0.05),与油酸相似(P>0.05)。结论:TMC60可较好地促进尼莫地平凝胶的体外透皮扩散。
Objective: To investigate the effect of N-trimethyl chitosan on the transdermal absorption of nimodipine gel. Methods: Nimodipine gel (TMC60) was prepared by using poloxamer 407 as gel matrix with N-trimethyl chitosan (TMC60) with a quaternization degree of 60% . In vitro transdermal studies were performed with a Franz diffusion cell and were compared with those of the azone (azone group), the nimodipine gel of the oleic acid (oleic acid group) and the nimodipine gel without a promoter (blank group ) Of the effect of comparison. Results: The steady state transdermal rate (J, μg · cm -2 · h -1) of blank group, azone group, TMC60 group and oleic acid group were 6.59, 13.22, 16.39 and 16.77, respectively. Compared with the blank group, TMC60 nimodipine gel had significant transdermal absorption promoting effect (P <0.05), and the effect intensity was better than azone (P <0.05), similar to oleic acid (P> 0.05). Conclusion: TMC60 can promote the in vitro transdermal diffusion of nimodipine gel.
