【目的】探讨Persephin对6鄄羟基多巴(6鄄OHDA)损伤神经干细胞的保护作用。【方法】用RT鄄PCR法从人胚胎脑获得PersephincDNA,构建重组Persephin腺病毒。用Westernblot方法分析Persephin基因的表达,Hoechst染色和流式细胞术测定6鄄羟基多巴对神经干细胞凋亡的诱导作用。【结果】从人胎脑成功克隆了人PersephincDNA,构建了其腺病毒载体。Westernblot证实重组Persephin腺病毒在神经干细胞中的表达,表达的Persephin可抑制6鄄羟基多巴诱导的神经干细胞凋亡。【结论】Persephin对6鄄羟基多巴损伤神经干细胞具有保护作用。
【Objective】 To investigate the protective effect of Persephin on neural stem cells injured by 6-hydroxydopamine (6-OHDA). 【Methods】 Persephinc DNA was obtained from human embryonic brain by RT-PCR and recombinant adenovirus was constructed. The expression of Persephin gene was analyzed by Western blotting. Hoechst staining and flow cytometry were used to detect the induction of neural stem cells apoptosis induced by 6-hydroxydopamine. 【Result】 Human Persephinc DNA was successfully cloned from human fetal brain and its adenoviral vector was constructed. Western Blot confirmed the expression of recombinant Persephin adenovirus in neural stem cells, and the expression of Persephin could inhibit the apoptosis of neural stem cells induced by 6-hydroxydopamine. 【Conclusion】 Persephin has a protective effect on neural stem cells damaged by 6-hydroxydopamine.
