G-MDSC在肿瘤发生中细胞丰度、周期及DNA合成的变化

目的:通过对C57BL/6J小鼠皮下接种Lewis lung carcinoma line细胞,探究荷瘤条件下粒细胞样髓源性抑制细胞(G-MDSC)的增殖变化。方法:用流式细胞术对骨髓细胞参数进行测定。结果:G-MDSC在荷瘤鼠骨髓中占总细胞数的百分比为(51.007±0.016)%,显著高于正常鼠骨髓中的百分比(12.696±0.004)%(P<0.01)。G-MDSC在荷瘤鼠中G0/G1期为(52.43±0.72)%,在正常鼠中为(56.73±1.50)%,差异无统计学意义(P=0.061);在荷瘤鼠中S期为(34.62±0.28)%,在正常鼠中为(31.82±0.88)%,差异有统计学意义(P<0.05);在荷瘤鼠中G2/M期为(14.92±0.71)%,在正常鼠中为(12.11±0.38)%,差异亦有统计学意义(P<0.05)。G-MDSC在荷瘤鼠中EdU阳性率为(57.35±0.35)%,显著高于正常鼠EdU阳性率(52.43±0.96)%(P<0.01)。结论:G-MDSC在荷瘤鼠中的增殖显著高于正常鼠。 OBJECTIVE: To investigate the proliferation of granulocyte-like myeloid derived suppressor cells (G-MDSCs) in tumor-bearing mice by inoculating Lewis lung carcinoma line cells into C57BL / 6J mice subcutaneously. Methods: Bone marrow cell parameters were determined by flow cytometry. Results: The percentage of G-MDSC in the bone marrow of tumor-bearing mice was (51.007 ± 0.016)%, which was significantly higher than that in normal mice (12.696 ± 0.004)% (P <0.01). G-MDSC in tumor-bearing mice G0 / G1 phase (52.43 ± 0.72)%, in normal mice (56.73 ± 1.50)%, the difference was not statistically significant (P = 0.061) (34.62 ± 0.28)% in normal mice, and (31.82 ± 0.88)% in normal rats, the difference was statistically significant (P <0.05); G2 / M phase was (14.92 ± 0.71)% in tumor- (12.11 ± 0.38)% in rats, the difference was also statistically significant (P <0.05). The positive rate of EdU in G-MDSC was (57.35 ± 0.35)%, which was significantly higher than that in normal mice (52.43 ± 0.96)% (P <0.01). Conclusion: The proliferation of G-MDSC in tumor-bearing mice was significantly higher than that in normal mice.
