为验证NAG微量酶反应比色法检测干扰素活性的实验条件,检测了27例癌症患者外周血单个核细胞conA诱生的γ干扰素。结果显示肝癌、肺癌、乳腺癌患者干扰素水平分别为52068±1124、40195±984、51282±1075,均显著低于对照组(280275±31321,P<001)。本法可检测至15×103细胞/孔,当活细胞数目以倍比稀释递减时,OD值也随之降低,细胞浓度与OD值基本呈一直线关系,有较高的敏感性和准确性;该方法比微量细胞病变抑制法稳定、准确,不受主观因素的影响
In order to verify the experimental conditions for the detection of interferon activity by NAG micro-enzyme reaction colorimetric assay, interferon-γ induced by conA in peripheral blood mononuclear cells of 27 cancer patients was tested. The results showed that the levels of interferon in liver cancer, lung cancer and breast cancer were 52068 ± 1124, 40195 ± 984, 51282 ± 1075, respectively, which were significantly lower than those in the control group (280275 ± 313 21, P <001). This method can be detected to 1 5 × 103 cells / hole, when the number of viable cells in a multiple dilution ratio decreased, OD value also decreases, the cell concentration and OD value was a linear relationship with high sensitivity and Accuracy; this method is more stable and accurate than the method of micro-cytopathic effect, not influenced by subjective factors
