目的:了解芪竹方对MGC803细胞血管内皮生长因子VEGF及其受体KDR表达的影响。方法:分成芪竹方高、低浓度、DMSO对照、空白对照4组处理MGC803细胞,运用RT-PCR、免疫荧光法研究VEGF、KDR的表达量。结果:芪竹方高、低浓度、DMSO对照、空白对照4组VEGFmRNA表达量光密度积分分别是:(6.7±2.12)、(9.98±3.27)、(18.99±7.73)、(20.55±4.85),芪竹方高浓度与空白对照组间差别具有显著性;KDRmRNA分别是(49.66±9.76)、(38.22±3.54)、(47.5±5.87)、(45.18±7.25),各组间无显著性差别;VEGF蛋白表达量荧光度值分别是:(24.6±5.5)、(29.4±4.8)、(43.2±7.4)、(40.4±3.2),芪竹方高浓度与空白对照组间差别具有显著性;KDR蛋白表达量荧光度值分别是:(32.2±8.6)、(35.4±8.3)、(37.4±11.3)、(27.4±4.7),各组间无显著性差别。结论:芪竹方对MGC803细胞VEGF表达具有下调作用,对KDR表达无明显影响。芪竹方抑制胃癌的作用可能与抑制VEGF表达有关。
Objective: To investigate the effect of Qi Zhu Zhu on the expression of vascular endothelial growth factor (VEGF) and its receptor KDR in MGC803 cells. METHODS: MGC803 cells were treated with Qizhuzhu high-dose, low-concentration, DMSO control, and blank control groups. The expression of VEGF and KDR were detected by RT-PCR and immunofluorescence. RESULTS: The optical density of VEGF mRNA expression in the four groups were high (6.7±2.12), (9.98±3.27), (18.99±7.73), and (20.55±4.85). There was a significant difference between the high concentration of Qizhufang and the blank control group; KDR mRNA was (49.66±9.76), (38.22±3.54), (47.5±5.87), (45.18±7.25), respectively, and there was no significant difference among the groups. The fluorescence values of VEGF protein expression were (24.6±5.5), (29.4±4.8), (43.2±7.4), and (40.4±3.2), respectively. There was a significant difference between the high concentration of Qizhufang and the blank control group; KDR The fluorometric values of protein expression were: (32.2±8.6), (35.4±8.3), (37.4±11.3), (27.4±4.7), and there was no significant difference among the groups. Conclusion: Qizhuzhu has a down-regulation effect on the expression of VEGF in MGC803 cells and has no significant effect on the expression of KDR. Qi Zhu Fang’s role in inhibiting gastric cancer may be related to the inhibition of VEGF expression.
