目的观察水溶性氮酮对体外培养的人眼小梁细胞增殖活性和超微结构的影响,寻找氮酮作用于眼部的安全浓度。方法进行人眼小梁细胞的体外培养,MTT法观察不同浓度氮酮作用不同时间后,对小梁细胞增殖的影响;倒置相差显微镜和透射电镜下观察氮酮对小梁细胞形态和超微结构的影响;荧光染色法观察氮酮对小梁细胞活性的影响。结果10-5~10-8g/L的氮酮作用24 h不影响小梁细胞的增殖和活性,10-4g/L的氮酮作用12 h不影响小梁细胞的增殖。10-6~10-8g/L氮酮作用24 h,10-5g/L氮酮作用12 h对人小梁细胞形态和超微结构无影响。荧光染色法显示10-3g/L和10-4g/L氮酮作用6 h后的细胞呈不同程度变性、坏死。结论10-6~10-8g/L浓度的氮酮不会影响小梁细胞的超微结构、增殖和活性,氮酮有望在眼科广泛应用。
Objective To observe the effect of water-soluble Azone on proliferation and ultrastructure of human trabecular meshwork cells cultured in vitro and find the safe concentration of Azone on the eye. Methods Human trabecular meshwork cells were cultured in vitro. The effects of different concentrations of azone on the proliferation of trabecular meshwork cells were observed by MTT assay. The morphology and ultrastructure of trabecular meshwork cells were observed under inverted phase contrast microscope and transmission electron microscope. The effect of Azone on trabecular cell activity was observed by fluorescence staining. Results Azone treated with 10-5 ~ 10-8 g / L did not affect the proliferation and activity of trabecular cells for 24 h. Azone at 10-4 g / L did not affect the proliferation of trabecular cells. The effects of 10-6 ~ 10-8g / L Azone for 24 h on the morphology and ultrastructure of human trabecular meshwork cells were observed after azone dose of 10-5g / L for 12 h. Fluorescent staining showed that cells of 10-3g / L and 10-4g / L Azone were denatured and necrotic to varying degrees after 6 h treatment. Conclusion Azone at a concentration of 10-6 ~ 10-8g / L does not affect the ultrastructure, proliferation and activity of trabecular cells. Azone is expected to be widely used in ophthalmology.
