目的探讨乙型肝炎病毒 x 蛋白(Hepatitis B virus X protein,HBx)和缺氧诱导因子-1α(Hypoxia-inducible factor,HIF-la)在肝癌组织中的表达及关系。方法采用免疫组织化学染色方法检测78例经福马林固定、石蜡包埋的原发性肝癌组织标本中HBx 和 HIF-1α的表达;免疫荧光检测 HepG2及稳定转染 HBx 基因的 HepG2(HBx-transfected HepG2)细胞中 HIF-1α的表达。结果78例肝癌组织标本中,HBx 和 HIF-1α免疫组织化学染色阳性率分别为74.23%(58/78)和69.23%(54/78);免疫荧光检测表明:正常氧状态下,HeptG2中 HIF-1α的表达阴性而 HBx-transfected HepG2中表达阳性,主要位于细胞浆.部分位于细胞核.在缺氧状态下,HeptG2和 HBx-transfected HepG2的细胞质和细胞核均有表达.结论 HBx 及 HIF-1α在人肝细胞肝癌组织中广泛表达,二者存在明显相关(P<0.01),在正常氧状态下,HBx 可诱导 HIF-α在 HepG2细胞中表达。提示:共同表达可能对原发性肝癌细胞的形成有重要作用。
Objective To investigate the expression and relationship of hepatitis B virus X protein (HBx) and hypoxia inducible factor-1α (HIF-1α) in hepatocellular carcinoma (HCC). Methods Immunohistochemical staining was used to detect the expression of HBx and HIF-1α in 78 specimens of primary hepatocellular carcinoma fixed by paraffin and paraffin-embedded. The expression of HBx and HIF-1α in HepG2 and HBx- transfected HepG2) cells. Results The positive rates of HBx and HIF-1α in 78 specimens of hepatocellular carcinoma were 74.23% (58/78) and 69.23% (54/78), respectively. The results of immunofluorescence showed that the positive expression of HIF -1α and negative in HBx-transfected HepG2 cells, mainly located in the cytoplasm, partly in the nucleus.Expression of both cytoplasm and nucleus in HeptG2 and HBx-transfected HepG2 cells was observed under hypoxia condition.Conclusion HBx and HIF-1α (P <0.01). Under normal oxygen conditions, HBx could induce the expression of HIF-α in HepG2 cells. Tip: Co-expression may play an important role in the formation of primary hepatoma cells.
