目的比较屏障系统内4种不同水处理方式的灭菌效果,为实验动物的饮水管理提供依据。方法从反渗透实验动物饮水、在线灭菌水、桶装纯净水、高压灭菌瓶装水及自来水分别取样接种培养,空白培养基为阴性对照,以培养基上菌落的生长情况作为评判屏障系统实验动物饮水的标准。最后选取一种除菌效果最好的方式在屏障内连续供水4 d,每天取剩余水样培养观察水中滋生细菌量与饲喂时间的关系。结果反渗透实验动物饮水和桶装纯净水采样后发现仍有少量菌落存在,但在线灭菌水与高压灭菌瓶装水均无菌落生长,最后选择在线灭菌水在屏障内被实验动物饮用1~4 d,每天检测发现菌落总数呈指数级增长。结论在线灭菌水与高温灭菌瓶装水可达到无菌要求,但是在饮用过程中存储时间越短越好,最佳方式则为当天灌喂。
Objective To compare the sterilizing effects of four different water treatment methods in the barrier system to provide basis for experimental animals drinking water management. Methods The samples were inoculated from drinking water, sterilized water, bottled purified water, autoclaved bottled water and tap water in reverse osmosis experimental animals respectively. Blank medium was taken as negative control, and the growth of colonies on culture medium was used as the evaluation barrier system experiment Animal drinking standards. Finally, choose the best way to remove bacteria in the barrier continuous water supply 4 d, take the remaining water samples every day to observe the relationship between breeding bacteria and feeding time in water. Results The results of reverse osmosis experimental animals drinking water and bottled pure water sampling and found that there are still a small amount of colonies exist, but the online sterilized water and autoclaved bottled water were no colony growth, and finally select the online sterilized water in the barrier was drinking experimental animals 1 ~ 4 days, the total number of colonies detected exponentially increased every day. Conclusions Online sterilizing water and autoclaved bottled water can reach sterility requirements, but the shorter the storage time is, the better the drinking process. The best way is to irrigate the same day.
