目的研究双氢青蒿素对前列腺癌PC-3细胞的凋亡诱导作用,并探讨其可能的机制。方法MTT法测定不同浓度双氢青蒿素对PC-3细胞增殖的影响,流式细胞仪(FCM)、透射电镜(TEM)观察双氢青蒿素对PC-3细胞的凋亡诱导作用,免疫组化(SP)检测双氢青蒿素对PC-3细胞内Bax、Bcl-2蛋白阳性表达的影响。结果双氢青蒿素对PC-3细胞有明显增殖抑制效应,能诱导PC-3细胞凋亡且具有浓度-时间依赖性,导致PC-3细胞线粒体肿胀、核碎裂、凋亡小体形成;随着双氢青蒿素浓度的增加,Bcl-2蛋白阳性表达降低,而Bax蛋白阳性表达增加。结论双氢青蒿素可能通过上调Bax,下调Bcl-2蛋白表达诱导前列腺癌PC-3细胞凋亡。
Objective To study the effect of dihydroartemisinin on the apoptosis of prostate cancer PC-3 cells and to explore its possible mechanism. Methods The effects of different concentrations of dihydroartemisinin on the proliferation of PC-3 cells were determined by MTT assay. The apoptosis induced by dihydroartemisinin on PC-3 cells was observed by flow cytometry (FCM) and transmission electron microscopy (TEM) The effect of dihydroartemisinin on the positive expression of Bax and Bcl-2 protein in PC-3 cells was detected by immunohistochemistry (SP). Results Dihydroartemisinin could significantly inhibit the proliferation of PC-3 cells and induce the apoptosis of PC-3 cells in a time-and concentration-dependent manner, resulting in mitochondrial swelling, nuclear fragmentation and formation of apoptotic bodies in PC-3 cells ; With the increase of dihydroartemisinin concentration, Bcl-2 protein expression decreased, while Bax protein expression increased. Conclusion Dihydroartemisinin may induce apoptosis of PC-3 cells by up-regulating Bax and down-regulating Bcl-2 protein expression.
