目的研究金丝桃苷(Hyp)对小鼠乙醇性胃粘膜损伤的保护作用及其与氧自由基(OFR)、一氧化氮(NO)和前列腺素(PG)的关系。方法制备小鼠乙醇性胃粘膜损伤模型,测定胃粘膜损伤面积;测定血浆、胃组织内MDA、NO和PGE2含量。结果Hyp(20、40、80mg·kg-1,ip)可剂量依赖性地抑制无水乙醇诱发的小鼠胃粘膜损伤,其损伤抑制率分别为389%、496%和556%。Hyp可使小鼠胃粘膜损伤过程中血浆、胃组织中升高的MDA含量降低,使降低的NO水平回升。Hyp对血浆、胃组织中PGE2含量无明显影响。结论Hyp对小鼠乙醇性胃粘膜损伤具有保护作用,其作用机制与Hyp抗氧化作用和促进NO水平恢复正常有关。
Objective To study the protective effect of hyperin on alcohol induced gastric mucosal injury in mice and its relationship with oxygen free radicals (OFR), nitric oxide (NO) and prostaglandin (PG). Methods The model of alcoholic gastric mucosa injury in mice was prepared and the area of gastric mucosal lesion was determined. The contents of MDA, NO and PGE2 in plasma and gastric tissue were measured. Results Hyp (20, 40, 80 mg · kg-1, ip) inhibited gastric mucosal injury induced by ethanol in a dose-dependent manner in 38.9%, 49.6% and 555%, respectively 6%. Hyp reduces the elevated MDA level in plasma and gastric tissue during gastric mucosal injury in mice, resulting in a decrease in NO levels. Hyp on plasma, gastric tissue PGE2 content had no significant effect. Conclusions Hyp protects against alcohol-induced gastric mucosal injury in mice, and its mechanism of action is related to the anti-oxidative effect of Hyp and the normalization of NO levels.
