目的:探讨凋亡基因bcl-2与凋亡信号受体FasR在胶质瘤中的表达以及与胶质瘤的发生和治疗的关系。方法:采用流式细胞术方法检测8例正常脑组织及36例胶质瘤组织中Bcl-2蛋白及FasR的表达水平。结果:正常脑组织中无Bcl-2蛋白及FasR的阳性表达,而胶质瘤组织中Bcl-2蛋白的阳性表达率为38.89%(14/36),FasR的阳性表达率为75%(27/36)。与低度恶性胶质瘤(WHOⅠ-Ⅱ级)相比,高度恶性胶质瘤(WHOⅢ-Ⅳ级)中Bcl-2蛋白的阳性表达率明显升高(P<0.05)。结论:胶质瘤中高表达Bcl-2蛋白及FasR,提示Bcl-2基因过度表达与胶质瘤的发生有关,并为Fas配体或Fas单克隆抗体治疗胶质瘤提供参考。
Objective: To investigate the expression of apoptosis-related gene bcl-2 and FasR in gliomas and its relationship with the occurrence and treatment of gliomas. Methods: The expression of Bcl-2 protein and FasR in 8 normal brain tissues and 36 glioma tissues were detected by flow cytometry. Results: There was no positive expression of Bcl-2 protein and FasR in normal brain tissue, while the positive rate of Bcl-2 protein in glioma was 38.89% (14/36) and that of FasR was 75% (27 / 36). The positive rate of Bcl-2 protein in patients with high grade gliomas (WHOⅢ-Ⅳ grade) was significantly higher than that in the patients with low grade glioma (WHOⅠ-Ⅱ grade) (P <0.05). Conclusion: The overexpression of Bcl-2 protein and FasR in gliomas suggest that overexpression of Bcl-2 gene is involved in the pathogenesis of gliomas and provide a reference for the treatment of gliomas with Fas ligand or Fas monoclonal antibody.
