为探讨培养基和外植体对杜仲愈伤组织诱导率的影响,将子叶、茎和真叶(代号为B1、B2、B3)三种外植体分别接种于MS、B5、White、Miller、H、(代号分别为A1、A2、A3、A4、A5)5种培养基上,调查并分析各组合的愈伤组织诱导率。经F测验,培养基、外植体和培养基×外植体的互作,效果均极显著。经SSR测验,H、B5、MS三种培养基愈伤组织诱导率极显著高于其它两种培养基。外植体中以茎的诱导率最高。不同组合间以A2B3、A5B3、A5B1、A2B2、A5B2、A1B1、A1B17个组合的愈伤组织诱导率较高,达90%以上。
In order to investigate the effect of medium and explants on the callus induction rate of Eucommia ulmoides Oliver, three explants of cotyledon, stem and leaf (code B1, B2, B3) were inoculated into MS, B5, White, Miller, H, respectively (code-named A1, A2, A3, A4, A5) on the five kinds of medium, to investigate and analyze the callus induction rate of each combination. The F test, medium, explants and medium × explants interaction, the effect is extremely significant. The SSR test, H, B5, MS three callus induction rate was significantly higher than the other two media. The explants with the highest induction rate of stem. The combinations of A2B3, A5B3, A5B1, A2B2, A5B2, A1B1 and A1B were higher callus induction rate of more than 90%.
