研究糖尿病大鼠肾脏血管紧张素Ⅱ 2型受体及Ⅳ型胶原的mRNA表达。方法 :大鼠随机分成两组 :单肾切组、糖尿病组。实验第 8周 ,应用RT -PCR检测大鼠肾皮质血管紧张素Ⅱ 2型受体 (AT2 )及Ⅳ型胶原mRNA表达。同时用放免法检测大鼠肾皮质血管紧张素Ⅱ含量。结果 :糖尿病组大鼠尿蛋白排泄率 (t=9.32 ,P <0 .0 1)、肾皮质血管紧张素Ⅱ水平 (t=2 .2 7,P <0 .0 5 )及Ⅳ型胶原mRNA表达 (t =3.71,P <0 .0 1)均较单肾切组明显升高 ;而AT2 的mRNA水平却下降 (t=2 .35 ,P <0 .0 5 )。结论 :糖尿病肾组织AT2 受体mRNA表达的下降及Ⅳ型胶原表达的增加参与了糖尿病肾病的发生、发展。
To investigate the mRNA expression of angiotensin Ⅱ type 2 receptor and type Ⅳ collagen in diabetic rats. Methods: The rats were randomly divided into two groups: single nephrectomy group, diabetic group. At the 8th week of the experiment, the mRNA expression of AT2 and type Ⅳ collagen in rat renal cortex was detected by RT-PCR. At the same time, radioimmunoassay was used to detect the content of angiotensin Ⅱ in rat renal cortex. Results: Urinary protein excretion rate (t = 9.32, P <0.01), renal angiotensin Ⅱ level (t = 2.27, P <0.05) (T = 3.71, P <0.01) were significantly higher than those in the nephrectomy group, while the mRNA level of AT2 was decreased (t = 2.35, P <0.05). Conclusion: The decreased expression of AT2 receptor mRNA and the increased expression of collagen type Ⅳ in diabetic nephropathy are involved in the occurrence and development of diabetic nephropathy.
