目的研究胰腺癌组织中缺氧诱导因子2(HIF-2α)的表达及其与血管内皮生长因子(VEGF)、微血管密度(MVD)的关系,分析胰腺癌中HIF-2α表达促进肿瘤血管生成的作用及与临床病理特征之间的关系。方法应用免疫组化SP法检测60例胰腺癌及相应正常胰腺组织中HIF-2α、VEGF蛋白的表达和分布,计数MVD,分析其间的相关性及其与肿瘤临床病理特征之间的关系。结果HIF-2α和VEGF在胰腺癌组织中的表达均明显高于正常胰腺组织,胰腺癌组织中MVD值明显高于正常胰腺组织,差异均有统计学意义(P<0.05)。HIF-2α的表达与VEGF的表达及MVD值成正相关(P<0.05),HIF-2α与VEGF的表达与胰腺癌TNM分期、肿瘤大小有关。结论胰腺癌组织中HIF-2α呈过量表达,并与VEGF及MVD显著相关;它可能通过上调VEGF表达促进胰腺癌血管生成,进而影响胰腺癌的发生、发展。
Objective To study the relationship between the expression of hypoxia inducible factor 2 (HIF-2α) and vascular endothelial growth factor (VEGF) and microvessel density (MVD) in pancreatic cancer and to analyze the relationship between the expression of HIF-2α and angiogenesis in pancreatic cancer Role and the relationship between clinicopathological features. Methods Immunohistochemical SP method was used to detect the expression and distribution of HIF-2α and VEGF in 60 cases of pancreatic cancer and corresponding normal pancreas. MVD was counted to analyze the relationship between them and its clinicopathological features. Results The expression of HIF-2α and VEGF in pancreatic cancer tissues were significantly higher than that in normal pancreatic tissues. The MVD in pancreatic cancer tissues was significantly higher than that in normal pancreatic tissues (P <0.05). The expression of HIF-2α was positively correlated with the expression of VEGF and the MVD (P <0.05). The expression of HIF-2α and VEGF was related to TNM stage and tumor size of pancreatic cancer. Conclusions HIF-2α is overexpressed in pancreatic cancer tissues and is significantly associated with VEGF and MVD. It may promote the angiogenesis of pancreatic cancer by up-regulating the expression of VEGF, thereby affecting the occurrence and development of pancreatic cancer.
