目的 探讨膀胱癌细胞系中核转录因子 kB(NF kB)及其调控基因的表达以及药物刺激对其的影响。方法 免疫细胞化学检测 p65、p50、p52、c Rel、RelB、IkBα、增殖细胞核抗原(PCNA)的蛋白表达水平 ,逆转录 聚合酶链反应 (RT PCR)检测 p65、p50、p52、c Rel、RelB、IkBα、CyclinD1、白细胞介素 (IL) 8、PCNA的mRNA水平。采用人类肿瘤坏死因子 α(TNF α,2 0 μg/L)刺激细胞观察检测指标的变化。结果 在EJ、T2 4细胞系中均见p65、p50、p52、c Rel、RelB、IkBα、PC NA等的蛋白和mRNA表达 ,以及CyclinD1、IL 8的mRNA表达 ,TNF α(2 0 μg/L)刺激后 ,NF kB家族成员的核易位增加 ,各指标的mRNA表达水平也有不同程度的增强。结论 膀胱癌细胞系中可见NF kB家族的表达 ,TNF α刺激后其调控基因的表达增强
Objective To investigate the expression of nuclear factor kB (NF kB) and its regulatory genes in bladder cancer cell lines and the effects of drug stimulation on them. Methods Immunocytochemistry was used to detect the protein expression levels of p65, p50, p52, c Rel, RelB, IkBα and PCNA. The expression of p65, p50, p52, c Rel and RelB were detected by reverse transcriptase polymerase chain reaction , IkBα, CyclinD1, Interleukin (IL) 8, PCNA mRNA levels. Human tumor necrosis factor α (TNF α, 20 μg / L) stimulated cells to observe the changes of test indicators. Results The mRNA and protein expressions of p65, p50, p52, c Rel, RelB, IkBα and PCNA, and the mRNA expressions of CyclinD1 and IL8 in EJ and T2 4 cell lines were all significantly decreased. The levels of TNFα (20 μg / L ) Stimulation, NF kB family member nuclear translocation increased, the mRNA expression of each indicator also have varying degrees of enhancement. Conclusion The expression of NF kB family can be seen in bladder cancer cell lines, and the expression of its regulatory genes is enhanced after TNFα stimulation
