目的:观察补肾法、疏肝法对小鼠卵母细胞生长分化因子-9(growth differerntiation factor-9,GDF-9)蛋白及mRMA表达的影响。方法:将300只小鼠随机分为6组:补肾高、低剂量组;疏肝高、低剂量组;控制性超促排卵(COH)组和空白对照组,每组50只,应用Western blotting和Real-time PCR方法分别测定卵母细胞中GDF-9蛋白和mRNA的表达,并对比各组小鼠卵母细胞数量及优质卵泡率。结果:各治疗组卵母细胞中GDF-9蛋白及mRNA表达显著高于空白对照组及COH组,其中高剂量组优于低剂量组(P<0.05),COH组的GDF-9蛋白及mRNA表达较空白对照组降低(P<0.05);各治疗组及COH组卵母细胞数量高于空白对照组(P<0.05),治疗组中高剂量组卵母细胞数量高于低剂量组(P<0.05);各治疗组及空白对照组优质卵泡率优于COH组(P<0.05),治疗组中高剂量组的优质卵泡率优于空白对照组(P<0.05)。结论:补肾调经方、逍遥丸在促性腺激素应用的基础上能增加卵母细胞数量及优质卵泡率,其机制可能与调控卵母细胞中GDF-9的蛋白及mRNA表达有关。
Objective: To observe the effects of tonifying kidney and dispersing liver on the expression of growth-promoting factor-9 (GDF-9) and mRMA in mouse oocytes. Methods: 300 mice were randomly divided into 6 groups: high and low doses of kidney; high and low doses of Shugan; control group, 50 cases in control group and 50 cases in control group. Western blotting The expression of GDF-9 protein and mRNA in oocytes were detected by Real-time PCR and the number of oocytes and the rate of high-quality follicles in each group were compared. Results: The expression of GDF-9 protein and mRNA in oocytes of each treatment group was significantly higher than that of blank control group and COH group, especially in high-dose group and low-dose group (P <0.05). The GDF-9 protein and mRNA in COH group The number of oocytes in treatment group and COH group was higher than that in blank control group (P <0.05), and the number of oocytes in high-dose group was higher than that in low-dose group (P < 0.05). The rate of high-quality follicles in the treatment group and the blank control group was better than that in the COH group (P <0.05). The high-dose follicle rate was higher in the middle-high dose group than in the blank control group (P <0.05). Conclusion: Bushen Tiaojing Recipe and Xiaoyao Pill can increase the number of oocytes and the rate of high-quality follicles on the basis of gonadotrophin application. The mechanism may be related to the regulation of GDF-9 protein and mRNA expression in oocytes.
