利用聚乳酸 ( PLA) /聚乙二醇 ( PEG) /聚乳酸 ( PLA)三嵌段共聚物 ( PELA)和乳化溶剂提取技术制备了包裹胰岛素的生物降解纳米微球 ,研究了囊材组成和分散剂对微球结构及胰岛素包封率、固载量和释放速率的影响 ,优化了制备微球的条件 .透射电镜和动态光散射测试表明 ,微球大小均匀 ,皆呈规整球形 ;粒径分布范围为 1 0 0~ 2 0 0 nm.微球对胰岛素的包封率和固载量分别达 87.6%和 1 7.5% .体外释放实验表明 ,微球可持续释药 1 0 d左右
Insulin-containing biodegradable nanospheres were prepared using polylactic acid (PLA) / polyethylene glycol (PEG) / polylactic acid (PLA) triblock copolymer (PELA) and emulsified solvent extraction technology. Dispersant on microsphere structure and insulin encapsulation efficiency, solid load and release rate, optimizing the conditions for the preparation of microspheres.Transmission electron microscopy and dynamic light scattering test showed that the microspheres were uniform size, are regular spherical; particle size Their distribution ranged from 100 nm to 200 nm.The entrapment efficiency and immobilization capacity of the microspheres to insulin were 87.6% and 7.5% respectively.The in vitro release experiments showed that the microspheres could be sustainedly released for about 10 days
