柱前衍生化HPLC-UV法检测桑叶中1-脱氧野尻霉素含量

目的建立芴甲氧羰酰氯柱前衍生化HPLC-UV法检测桑叶中1-脱氧野尻霉素(DNJ)含量的方法。方法采用HPLC-UV法检测,以芴甲氧羰酰氯为衍生化试剂,色谱柱为Hedera ODS-2 C_(18)柱(4.6×250 mm,5μm);以乙腈和0.1%磷酸水溶液作为流动相,梯度洗脱;流速为1.0 m L/min;检测波长为254 nm;柱温30℃。结果 DNJ在0.0046~0.2960 mg/m L范围内线性关系良好(R2=0.9999),平均回收率为99.33%,RSD为2.75%(n=9)。结论方法快速、简便、准确,适用于桑叶中DNJ的定量分析。 OBJECTIVE To establish a method for the determination of 1-deoxynojirimycin (DNJ) in mulberry leaves by HPLC-UV with fluorene methoxycarbonyl chloride pre-column derivatization. Methods The HPLC-UV method was used. Fluorescent methoxycarbonyl chloride was used as the derivatization reagent. The column was Hedera ODS-2 C 18 column (4.6 × 250 mm, 5 μm). Acetonitrile and 0.1% phosphoric acid aqueous solution were used as the mobile phase , Gradient elution; flow rate of 1.0 m L / min; detection wavelength of 254 nm; column temperature 30 ℃. Results DNJ had a good linearity (R2 = 0.9999) in the range of 0.0046-0.2960 mg / mL with an average recovery of 99.33% and RSD of 2.75% (n = 9). Conclusion The method is rapid, simple and accurate and is suitable for the quantitative analysis of DNJ in mulberry leaves.
