目的 探讨分化诱导剂六亚甲基二乙酰胺 (hexamethylenebisacetamide ,HMBA)对人粘液表皮样癌细胞系MEC 1细胞增殖和凋亡相关基因表达的影响。方法 用 0 0 0 2mol/LHMBA对培养的MEC 1细胞进行体外诱导 72h后 ,分别采用光镜、免疫组化、原位杂交、图像分析等方法进行增殖细胞核抗原 (proliferatingcellnuclear,PCNA)、bax、bcl 2蛋白及mRNA表达水平的检测。结果 经HMBA诱导的MEC 1细胞中PCNA的表达较对照组明显下降 (P <0 0 1) ,同时MEC 1细胞的凋亡相关基因bax、bcl 2蛋白及mRNA表达均明显上升 (P <0 0 1)。结论 HMBA对MEC 1细胞的增殖基因PCNA有明显的负调控作用 ,从而可抑制其增殖 ;并且它能够特异性上调bax和bcl 2基因的表达 ,发挥了促进凋亡的作用。
Objective To investigate the effect of differentiation-inducing agent hexamethylenebisacetamide (HMBA) on proliferation and apoptosis-related gene expression in human mucoepidermoid carcinoma cell line MEC 1. Methods The cultured MEC 1 cells were induced with 0 0 0 2mol / LHMBA for 72 h in vitro. The proliferating cell nuclear antigen (PCNA), bax, bcl 2 protein and mRNA expression levels were detected. Results The expression of PCNA in MEC-1 cells induced by HMBA was significantly lower than that in control group (P <0.01), and the protein and mRNA expression of bax, bcl-2 in MEC 1 cells were significantly increased (P <0 0 1). CONCLUSION: HMBA can negatively regulate the proliferation of PC-1 cells and inhibit the proliferation of MEC-1 cells. In addition, it can specifically up-regulate the expression of bax and bcl-2 genes and promote apoptosis.
