通过血凝试验及糖抑制试验,测定褶纹冠蚌的血清及肌肉提取液的凝血活性及糖抑制专一性,同时进行热稳定性、pH及Ca2+影响试验。结果表明,血清和肌肉凝集素都可以凝集多种脊椎动物的红细胞,其中对鸡红细胞的凝集效价均为最高,分别为25和21;血清和肌肉凝集素都不能凝集大肠杆菌,均能凝集嗜水气单胞菌、枯草杆菌、变形杆菌和保加利亚乳杆菌。血清和肌肉凝集素都有一定的热稳定性,血清凝集素60℃处理10min丧失凝集活性,而肌肉凝集素50℃处理10min凝集活性丧失。血清凝集素具有较广泛的pH适应性,在pH6~10内对鸡红细胞均有凝集活力,而肌肉提取液凝集素在pH<7或pH>9时凝集活性消失。血清凝集素能被乳糖和麦芽糖所抑制,肌肉凝集素不能被受试的7种糖所抑制。血清凝集素凝集活性明显地依赖于Ca2+等2价离子,而肌肉凝集素对Ca2+等2价离子没有依赖性。因此,认为褶纹冠蚌血清和肌肉提取液所含凝集素类型不同。用嗜水气单胞菌对褶纹冠蚌进行免疫刺激后,血清和肌肉凝集素对鸡红细胞的凝集效价并没有发生明显变化。
Through the hemagglutination test and the sugar inhibition test, the coagulation activity and specificity of sugar inhibition of the serum and muscle extracts of Plicata chinensis were determined, and the effects of heat stability, pH and Ca2 + were also tested. The results showed that serum and lectin can agglutinate red blood cells of a variety of vertebrates, of which the highest agglutination titers for chicken erythrocytes were 25 and 21, respectively; neither serum nor lectin agglutinated E. coli, both agglutinated Aeromonas hydrophila, Bacillus subtilis, Proteus and Lactobacillus bulgaricus. Serum and lectin have certain thermal stability, agglutination of lectin at 60 ℃ for 10min loses the agglutination activity, and lectin agglutinin 50 ℃ 10min cohesion activity loss. Serum lectin has a wide range of pH adaptability, agglutination activity of chicken erythrocytes at pH 6 ~ 10, and the lectin of muscle extract disappears when pH <7 or pH> 9. Serum lectin can be inhibited by lactose and maltose, and the lectin can not be inhibited by the seven sugars tested. The agglutination activity of serum lectin obviously depends on the divalent ions such as Ca2 +, whereas the lectin does not depend on the divalent ions such as Ca2 +. Therefore, it is thought that the types of lectins contained in the serum and muscle extracts of C. plicata are different. Agglutination titer of serum and lectin on chicken erythrocytes did not change significantly after immunostimulation with Aeromonas hydrophila on Plica przewalskii.
