目的:利用生物信息技术,对丙型肝炎病毒NS5区抗原决定簇进行预测分析,获得具有良好抗原的重组蛋白。方法:以Goldkey程序,通过综合亲水参数、可及性参数、柔韧性参数、抗原性参数、氨基酸序列的电荷分布和β折叠等多种指标,对HCVNS5区抗原决定簇进行预测分析,克隆并表达部分NS5区基因。结果:发现7个可能性最大的抗原决定簇肽段,在此基础上表达的NS5区融合蛋白可与丙型肝炎病人血清发生特异性反应。结论:根据Goldkey程序的预测结果,获得了具有良好抗原性的HCVNS5重组蛋白
OBJECTIVE: To predict and predict the antigenic determinants of hepatitis C virus NS5 region by using bioinformatics technology and obtain the recombinant protein with good antigen. Methods: The Goldkey program was used to predict the antigenic determinants of HCVNS5 region through comprehensive combination of hydrophilicity parameters, accessibility parameters, flexibility parameters, antigenicity parameters, amino acid sequence charge distribution and β-sheet. On the basis of this, the NS5 fusion protein expressed in the serum could specifically react with the serum of hepatitis C patients.
