采用农杆菌介导玉米萌发种子转化方法,将海藻糖合酶基因(TPS)导入玉米自交系昌7-2中。PCR和Southern blot检测结果表明,目的基因已导入转化植株并整合到受体基因组上。田间抗旱性鉴定和生理生化指标结果显示,转基因植株(株系)的抗旱性明显高于对照。说明海藻糖合酶基因具有抗旱功能,利用植物基因工程技术对其进行玉米抗旱育种是切实可行的。
Agrobacterium-mediated maize seed germination transformation method, the trehalose synthase gene (TPS) into maize inbred line Chang 7-2. PCR and Southern blot results showed that the target gene was introduced into the transformed plant and integrated into the receptor genome. Field drought resistance identification and physiological and biochemical indicators showed that the transgenic plants (lines) drought resistance was significantly higher than the control. It indicates that the trehalose synthase gene possesses drought resistance function and it is practicable to use plant gene engineering technology to carry out drought resistant breeding of corn.
