为研究雄激素致前列腺良性增生的分子机理,分离培养了人胎儿前列腺间质细胞,并给予DHT(1~10000μg/L)刺激,MTT法测定。结果显示,DHT对该细胞具有刺激增殖作用,其最适刺激浓度为1000μg/L。斑点杂交结果显示,鸟氨酸脱羧酶(ODC)mRNA的表达在DHT(1000μg/L)刺激6h时开始升高,24h时达高峰,30h时有所降低。提示雄激素刺激前列腺间质细胞增殖的分子机理部分是通过刺激前列腺间质细胞的ODC基因表达来完成的。
To study the molecular mechanism of androgen-induced benign prostatic hyperplasia, human fetal prostatic stromal cells were isolated and cultured and stimulated with DHT (1 ~ 10000 μg / L) for MTT assay. The results showed that DHT on the cells have stimulated proliferation, the optimum stimulus concentration of 1000μg / L. Dot hybridization showed that the expression of ornithine decarboxylase (ODC) mRNA began to increase at 6h after stimulation with DHT (1000μg / L), reached peak at 24h and decreased at 30h. It is suggested that the molecular mechanism of androgen stimulation of prostate stromal cell proliferation is partly achieved by stimulating the expression of ODC gene in stromal cells of prostate.
