目的从分子水平探讨男性生殖系感染大肠埃希菌后导致不育的机理,精浆活性氧的含量与肿瘤坏死因子(TNF-α)、白介素-6(IL-6)、白介素-10(IL-10)和白介素18(IL-18)的相互关系及其在不育中的作用。方法选择60例大肠埃希菌感染男性不育患者作为大肠埃希菌感染不育组、30例无感染不育患者和健康已育男性30例分别作为无感染不育对照组和健康对照组,采用硫代巴比妥酸比色法测定精浆丙二醛(MDA)含量,黄嘌呤氧化酶法检测超氧化物歧化酶(SOD)活性,酶联免疫吸附法测定IL-6、IL-10、IL-18和TNF-α的水平,分析MDA与IL-10、IL-18和TNF-α的相关性。结果大肠埃希菌感染男性不育患者精浆MDA含量、IL-6、IL-18和TNF-α含量均高于无感染不育组和健康对照组(P<0.01),IL-10含量和SOD活性下降;精浆MDA含量与TNF-α(r=0.67)和IL-18(r=0.51)呈正相关,与IL-10呈负相关(r=-0.54)。结论生殖系统大肠埃希菌感染所致男性不育与体内活性氧增加、抗氧化物质降低、细胞因子间网络平衡紊乱有关。
Objective To investigate the mechanism of male infertility induced by Escherichia coli infection in male reproductive system at molecular level. The content of reactive oxygen species in seminal plasma was positively correlated with tumor necrosis factor (TNF-α), interleukin-6 (IL-6), interleukin-10 -10) and interleukin 18 (IL-18) and their role in infertility. Methods Sixty infertile men with infective Escherichia coli were enrolled as infertility group of Escherichia coli, 30 infertile infertile men and 30 healthy male infertile men as non-infertile control group and healthy control group, respectively. The content of malondialdehyde (MDA) in seminal plasma was determined by thiobarbituric acid colorimetric assay. The activity of superoxide dismutase (SOD) was detected by xanthine oxidase method. The levels of IL-6 and IL-10 , IL-18 and TNF-α, and to analyze the correlation between MDA and IL-10, IL-18 and TNF-α. Results The levels of MDA, IL-6, IL-18 and TNF-α in the seminal plasma of infertile Escherichia coli group were higher than those in the infertile group and healthy control group (P <0.01) The activity of SOD in seminal plasma was positively correlated with TNF-α (r = 0.67) and IL-18 (r = 0.51), but negatively correlated with IL-10 (r = -0.54). Conclusion The infertility caused by Escherichia coli in the reproductive system is related to the increase of reactive oxygen species and antioxidants in the male and the imbalance of cytokines in the network.
