目的探讨 Apaf-1在大肠癌中的表达及其与Caspase-3及Ki-67的相关性。方法收集大肠癌(肠癌组)、腺瘤(腺瘤组)及正常黏膜组织(正常组)标本,每组各100例,免疫组化 SP 染色方法检测 Apaf-1表达,并分析其与Caspase-3、Ki-67相关性。结果肠癌组、腺瘤组及正常组中,Apaf-1蛋白的阳性表达率分别为39.00%、88.00%、95.00%;Caspase-3蛋白的阳性表达率分别为45.00%、83.00%、91.00%,肠癌组中 Apaf-1、Caspase-3阳性率低于腺瘤组及正常组(P <0.05);Ki-67蛋白的阳性表达率分别为68.00%、26.00%、8.00%,肠癌组中Ki-67阳性率高于腺瘤组及正常组(P<0.05)。Apaf-1与Caspase-3阳性表达呈正相关(r=0.345,P <0.01),与Ki-67阳性表达呈负相关(r=-0.361,P <0.01)。结论 Apaf-1、Caspase-3、Ki-67均参与了大肠癌的发展过程,推测大肠癌患者 Apaf-1受到抑制,进而Caspase-3活化减少,肿瘤细胞增加。“,”Obj ective]To explore the expression of apoptosis factor Apaf-1 in large intestinal cancer and its relationship with Caspase-3 and Ki-67.[Methods]The specimens of 100 cases of large intestinal cancer(intestinal cancer group),100 cases of adenoma(adenoma group)and 100 cases of normal mucosa(normal group)were col-lected.Immunohistochemical SP method was used to detect the expression of Apaf-1 .The relationship of the ex-pression of Apaf-1 with Caspase-3 and Ki-67 was analyzed.[Results]The positive expression rates of Apa-1 pro-tein in intestinal cancer group,adenoma group and normal group were 39.00%,88.00% and 95.00% respective-ly,while the positive expression rates of Caspase-3 protein were 45.00%,83.00% and 91.00%,respectively. The positive expression rates of Apaf-1 and Caspase-3 in intestinal tumor group were lower than those in adenoma group and normal group(P<0.05).The positive expression rates of Ki-67 protein in intestinal cancer group,ad-enoma group and normal group were 68.00%,26.00% and 8.00% respectively,and that in intestinal cancer group was higher than those in adenoma group and normal group(P <0.05).The positive expression of Apaf-1 was positively correlated with the positive expression of Caspase-3(r=0.345,P<0.01),and negatively correla-ted with the positive expression of Ki-67(r=-0.361,P<0.01).[Conclusion]Apaf-1,Caspase-3 and Ki-67 in-volve in the development of large intestinal cancer which infers that Apaf-1 in patients with large intestinal cancer is inhibited,thus the activation of Caspase-3 is reduced and tumor cell is increased.
