目的测定肥胖患者皮屑中钠、钾离子浓度及皮肤组织钠、钾转运相关蛋白的表达,并探讨其临床意义。方法招募127例重庆地区自愿者,并以体质指数(BMI)和腰围(WC)值,将其分为肥胖、超重、健康组和腹型肥胖、健康组,刮取研究对象双前臂内侧皮屑,用消化液(1.5 mL×65%硝酸和0.5 mL×70%高氯酸)加热溶解后,使用火焰原子分光光度计检测溶液中钠、钾的含量;提取小鼠及人皮肤组织蛋白,用蛋白印迹法检测糖皮质激素受体、醛固酮受体、Na+-K+-ATP酶表达。结果在最适反应条件下,本方法的相对标准偏差在0.34~1.45,回收率为93.1%~107.2%;肥胖组与健康组相比,皮屑钠、钾值均有显著性差异(P=0.002,0.000),肥胖组与超重组比差异也有显著性(P=0.002,0.000),超重组与健康组比较均无差异。Western blot检测小鼠和人皮肤组织中糖皮质激素受体、醛固酮受体、Na+-K+-ATP酶3种蛋白均有表达。结论应用火焰原子分光光度计测量肥胖者皮屑钠、钾含量明显升高,可简易地反映临床肥胖患者组织电解质变化,钠、钾转运相关的3种蛋白在小鼠和人皮肤组织中均有表达。
Objective To determine the concentration of sodium and potassium in dander and the expression of sodium and potassium in skin tissue in obese patients and to investigate its clinical significance. Methods A total of 127 volunteers from Chongqing were enrolled and divided into obesity, overweight, healthy group and abdominal obesity, healthy group according to body mass index (BMI) and waist circumference (WC) After digestion solution (1.5 mL × 65% nitric acid and 0.5 mL × 70% perchloric acid) was heated and dissolved, the content of sodium and potassium in the solution was detected by flame atomic spectrophotometer. The mouse and human skin tissue proteins were extracted with Western blotting was used to detect the expression of glucocorticoid receptor, aldosterone receptor and Na + -K + -ATPase. Results Under the optimal reaction conditions, the relative standard deviations (RSDs) of this method ranged from 0.34 to 1.45 and the recoveries ranged from 93.1% to 107.2%. The sodium and potassium values of dander in obesity group were significantly different from those in healthy group (P = 0.002,0.000). There was also significant difference between obesity group and overweight group (P = 0.002,0.000), there was no difference between overweight group and healthy group. Conclusion Flame atomic spectrophotometer (FAAS) can measure sodium and potassium in obese people easily, which can easily reflect the changes of tissue electrolytes in obese patients. The three proteins related to sodium and potassium transport in both obese and human skin tissues expression.
