目的:为探讨急性脑梗塞急性期患者血小板内游离钙、cAMP和TXB2含量之间的相互关系及意义。方法:用Fura—2/AM荧光技术测定了16例急性脑梗塞患者急性期血小板内钙含量,用放射免疫分析法测定其血小板内。AMP和血栓素B2(TXB2)含量。结果:患者血小板内游离Ca2+和TXB2含量明显高于正常对照(P<0.01),而血小板内cAMP含量则明显低于正常对照(P<0.01);血小板内游离Ca2+含量和TXB2含量与cAMP含量呈明显的负相关性(r分别为-0.771和-0.553,P<0.01),而血小板内Ca2+与TXB2成明显的正相关性(r为0.710,P<0.01)。结论:急性脑梗塞患者在急性期血小板功能处于活跃状态,可能与其内Ca2+含量增加、TXB2生成增多、cAMP含量降低有关,三者相互作用激活血小板。提示在急性脑梗塞急性期积极治疗的同时,使用抗血小板的药物有重要意义。
Objective: To investigate the relationship between the levels of intracellular free calcium, cAMP and TXB2 in patients with acute cerebral infarction and their significance. Methods: Platelet calcium levels were measured in 16 acute cerebral infarction patients by Fura-2 / AM fluorescence spectrophotometry. The platelets were determined by radioimmunoassay. AMP and thromboxane B2 (TXB2) content. Results: The content of free Ca2 + and TXB2 in platelet of patients was significantly higher than that of normal control (P <0.01), while the content of cAMP in platelet was significantly lower than that of normal control (P <0.01). The contents of free Ca2 + and TXB2 (R = -0.771 and -0.553, respectively, P <0.01), while there was a significant positive correlation between platelet Ca2 + and TXB2 (r = 0.710, P <0.01). Conclusion: In acute stage of acute cerebral infarction, platelet function is active, which may be related to its increase of Ca2 + content, TXB2 production and decrease of cAMP content. The interaction between them may activate platelet. Prompt acute acute cerebral infarction in the active treatment of the same time, the use of anti-platelet drugs is of great significance.
