目的建立HPLC法测定早产儿血清中茶碱的浓度,并将其应用于我院早产儿血药浓度监测。方法咖啡因作内标,血样经液-液萃取后,取有机相氮气吹干,甲醇复溶后进样测定,以Agilent Eclipse XDV-C_(18)柱(150 mm×4.6 mm,5μm)为分析柱,甲醇-水(22:78)为流动相,检测波长275 nm,流速1.0 mL·min~(-1)。结果茶碱在0.5~40 mg·L~(-1)(r=0.999 7)内线性关系良好,提取回收率>95%,日内、日间RSD均<5%。结论该法操作简便、快速,结果准确,适于我院早产儿血药浓度监测。
Objective To establish a HPLC method for the determination of serum theophylline concentration in preterm infants and to apply it in the monitoring of plasma concentrations of premature infants in our hospital. Methods Caffeine was used as internal standard. After the sample was extracted by liquid - liquid, the organic phase was air - dried with nitrogen and the sample was reconstituted with methanol. The sample was determined by Agilent Eclipse XDV - C 18 column (150 mm × 4.6 mm, 5 μm) The analytical column and methanol - water (22:78) were used as the mobile phase. The detection wavelength was 275 nm and the flow rate was 1.0 mL · min ~ (-1). Results The theophylline had a good linear relationship in the range of 0.5-40 mg · L -1 (r = 0.999 7). The recovery rate of theophylline was> 95%. The intra-day and inter-day RSD were all less than 5%. Conclusion The method is simple, rapid, accurate and suitable for the monitoring of blood concentration in preterm infants in our hospital.
