目的:研究黄连阿胶汤对阴虚火旺证模型大鼠尿儿茶酚胺(CAs)排泄量的影响。方法:给予大鼠腹腔注射促皮质素(ACTH,16 u/kg),每天1次,连续4 d,以复制大鼠阴虚火旺证模型;42只SD雄性大鼠随机均分为正常对照(等容蒸馏水)组、模型(等容蒸馏水)组、天王补心丸(1 g/100 g)组、首乌代鸡子黄(0.992 g/100 g)组和黄连阿胶汤高、中、低剂量(2.856、1.428、0.714 g/100 g)组,灌胃给药,每天1次,连续15 d。在给药第5、10、15天时称定大鼠体质量,收集24 h大鼠尿液并计量体积,采用高效液相色谱-荧光分析(HPLC-FD)法测定尿液中CAs含量。结果:与模型组和天王补心丸组比较,给药第5天,黄连阿胶汤高、中、低剂量组大鼠尿液中去甲肾上腺素(NE)、重酒石酸肾上腺素(E)、多巴胺(DA)含量减少,尿液体积(V)减少,差异均有统计学意义(P<0.05);给药第10天,黄连阿胶汤高、中、低剂量组大鼠尿液中NE、E含量减少,高、中剂量组尿液V减少,高、低剂量组大鼠尿液中DA含量减少,差异均有统计学意义(P<0.05);给药第15天,与天王补心丸组比较,黄连阿胶汤高、中、低剂量组大鼠尿液V和NE、E、DA含量减少,差异有统计学意义(P<0.05)。结论:阴虚火旺证模型大鼠对腹腔注射ACTH造成的尿液增多、尿CAs排泄量增高等病理改变具有一定的自愈能力;天王补心丸一定程度上抑制了模型大鼠的自愈;黄连阿胶汤能有效恢复阴虚火旺证模型大鼠的尿液V增多和尿CAs排泄量增高等病理改变,并且显示了良好的剂量依赖效应。
Objective: To investigate the effects of huanglian gelatin soup on urinary catecholamine (CAs) excretion in rats with yin deficiency and fire syndrome. Methods: Rats were injected intraperitoneally with corticotropin (ACTH, 16 u / kg) once a day for 4 days to replicate the model of Yin deficiency syndrome in rats. Forty-two SD male rats were randomly divided into normal control (1 g / 100 g), Radix Astragali Radix (0.992 g / 100 g) group, and berberine gelatin capsules high, medium and low dose (2.856,1.428,0.714 g / 100 g) group, intragastric administration, once a day for 15 days. Results: Compared with the model group and the Tiawang Buxin Xin Pill group, on the 5th day after administration, the contents of norepinephrine (NE), epinephrine bitartrate (E), norepinephrine Dopamine (DA), urine volume (V) decreased, the difference was statistically significant (P <0.05); on the 10th day after administration, the contents of NE, (P <0.05). On the 15th day after administration, the content of E decreased, while in the high and middle dose groups, the urine V decreased. In the high and low dose groups, the content of DA in the urine decreased significantly Compared with the bolus group, the contents of V, NE, E and DA in urine of high, medium and low dose groups of Huanglian-gelatin decoction decreased significantly (P <0.05). Conclusion: The Yin-Huo-Huo-Zheng rat model has some self-healing abilities to the pathological changes caused by intraperitoneal injection of ACTH, such as increased urinary CAs excretion; Huanglian donkey-hide gelatin can effectively restore pathological changes of urine V and excretion of urinary CAs in rats with Yin deficiency syndrome and show a good dose-dependent effect.
