水稻植株从外界吸收氮素越多,植株体内氨基态氮的含量也越多,因此,根据体内氨基态氮的含量可以知道外界的供氮水平及本身的需氮情况。几年来,我们用茚三酮法测定水稻植株体内氨基态氮的含量,取得了良好的效果。由于氨基态氮含有游离的氨基,它与水合茚三酮作用后产生蓝紫色的化合物,这种化合物颜色的深浅与氨基态氮的浓度成正比,因此,可以通过它们与标准色阶比较来确定氨基态氮的含量。测定的方法晴天,选取有代表性的水稻植株10株,分蘖期可取植株的全部叶片或叶鞘;穗分化后可取倒3—4叶的
