观察中药钙通道阻滞剂川芎嗪(TMP)和异博定(VP)对白血病HL60/HT耐药细胞系多药耐药(MDR)的逆转.方法:采用四唑氮蓝(MTT)法作为药物敏感实验,流式细胞仪测量细胞内柔红霉素(DNR)浓度.结果:采用非细胞毒剂量的100mg/LTMP和(或)0.32mg/L及1mg/L的VP与DNR联合应用,均能提高DNR的细胞杀伤作用,其效果比较为VP(1mg/L)+TMP>VP(1mg/L),VP(0.32mg/L)+TMP>TMP>VP(0.32mg/L)(P<0.01),而且VP(1mg/L)的逆转效果与VP(0.32mg/L)+TMP相当.同时用流式细胞仪测量细胞内DNR浓度发现TMP和(或)VP均能增加HL60/HT细胞内DNR的浓度,效果比较VP(1mg/L)+TMP>VP(1mg/L)>TMP.结论:TMP与VP有协同逆转作用,是一种有效的中药白血病MDR逆转剂.
To observe the reversal of the multidrug resistance (MDR) of leukemia HL60/HT-resistant cell line with traditional Chinese medicine calcium channel blockers TMP and VP. METHODS: Tetrazolium blue (MTT) method was used as drug sensitivity assay. Flow cytometry was used to measure the intracellular concentration of daunorubicin (DNR). RESULTS: The combination of non-cytotoxic doses of 100 mg/LTMP and/or 0.32 mg/L and 1 mg/L VP with DNR could increase the cytotoxicity of DNR. The effect was compared with VP (1 mg/L). + TMP> VP (1 mg/L), VP (0.32 mg/L) + TMP> TMP> VP (0.32 mg/L) (P<0.01), and the reversal effect of VP (1 mg/L) and VP ( 0.32mg/L) + TMP quite. At the same time, using flow cytometry to measure intracellular DNR concentration, it was found that both TMP and (or) VP could increase the concentration of DNR in HL60/HT cells. The effect was compared with VP (1 mg/L) + TMP> VP (1 mg/L)> TMP. Conclusion: TMP and VP have a synergistic reversal effect and it is an effective reversal agent for MDR leukemia.
