目的采用蛋白免疫印迹法(Western Blot)研究中药复方排毒止带汤含药血清对人宫颈鳞癌SiHa细胞人乳头瘤病毒16型(HPV16)E6,E7,p53,pRb蛋白表达的影响。探讨复方排毒止带汤对人乳头瘤病毒的影响。方法将大鼠随机分为空白对照组、25%含药血清组及50%含药血清组。空白对照组口服生理盐水,25%含药血清组及50%含药血清组口服复方排毒止带汤煎剂,将复方排毒止带汤25%含药血清和50%含药血清分别作用于SiHa细胞72,120 h后,采用Western Blot法检测HPV16 E6,E7,p53,pRb蛋白表达,采集数据。统计学分析采用独立样本t检验。结果复方排毒止带汤50%含药血清作用72 h,HPV16 E6,E7蛋白表达水平下降,p53,pRb蛋白表达水平升高,复方排毒止带汤50%含药血清作用120 h,HPV16 E6,E7蛋白表达水平较作用72 h明显下降,p53,Rb蛋白表达水平明显升高,2组相应指标比较P均<0.05,差异有统计学意义。复方排毒止带汤25%含药血清作用120 h,HPV16 E6,E7蛋白表达水平下降,p53,Rb蛋白表达水平上升,复方排毒止带汤50%含药血清作用120 h HPV16 E6,E7蛋白表达水平较25%含药血清明显下降,p53,Rb蛋白表达水平明显升高,2组相应指标比较P均<0.05,差异有统计学意义。结论复方排毒止带汤含药血清对HPV16 E6,E7,p53,Rb蛋白表达具有明显调控作用,复方排毒止带汤的活性具有一定的时效性。
Objective To investigate the effects of Chinese medicine compound detox cessation on serum E6, E7, p53 and pRb protein expressions of human papillomavirus 16 (HPV16) in human cervical squamous cell carcinoma by Western Blot. To explore the compound detoxification Zhitou Tang on human papillomavirus. Methods The rats were randomly divided into blank control group, 25% drug-containing serum group and 50% drug-containing serum group. Oral saline, 25% drug-containing serum group and 50% drug-containing serum group were given oral compound detoxification Zhidou Decoction. The compound detoxification Zhitou Decoction 25% drug-containing serum and 50% drug-containing serum were respectively applied to SiHa After 72 and 120 h, the protein expression of HPV16 E6, E7, p53 and pRb were detected by Western Blot and the data were collected. Statistical analysis using independent samples t test. The results showed that the expression of HPV16 E6 and E7 protein decreased and the expression of p53 and pRb protein increased after treated with 50% serum containing compound detoxification Zhitou Decoction for 72 h. The effect of 50% serum containing compound detoxification Zhidou Tang for 120 h, HPV16 E6, E7 protein expression was significantly lower than the role of 72 h, p53, Rb protein expression was significantly increased, the corresponding indicators of two groups were P <0.05, the difference was statistically significant. The serum level of HPV16 E6 and E7 decreased, while the expression of p53 and Rb protein increased, the expression of HPV16 E6 and E7 protein in compound detoxification Zhitou Decoction 50% serum for 120 h Compared with 25% serum, the level of p53 and Rb protein was significantly decreased, and the corresponding indexes of two groups were all P <0.05, the difference was statistically significant. Conclusion Compound detoxification Zhitai Tang containing serum on HPV16 E6, E7, p53, Rb protein expression has a significant regulatory effect, compound detoxification Zhitou Tang activity has some timeliness.
