目的通过在兔颈动脉球囊损伤模型中转染血小板衍化内皮细胞生长因子(PD-ECGF)基因使其上调,探讨该基因在预防血管再狭窄中的作用。方法 36只新西兰白兔(体质量2.5~3.0kg)建立兔颈动脉球囊损伤模型,分为实验组(n=18)和对照组(n=18):实验组使用病毒进行PD-ECGF基因转染;对照组采用交付剂处理。3周后,采用Evans蓝染色和HE染色分别检测各组内皮覆盖率及新生内膜的改变情况;采用免疫组化技术和免疫印迹法检测颈动脉组织中PD-ECGF蛋白表达情况。结果实验组内皮覆盖率[(72.8±4.0)%]显著高于对照组[(33.6±7.4)%];HE染色可见实验组内膜及中层平滑肌增生程度较对照组明显减轻;免疫组化及免疫印迹法结果均表明实验组颈动脉组织中PD-ECGF蛋白表达显著高于对照组(P<0.05)。结论在家兔颈动脉球囊损伤模型中,上调PD-ECGF基因具有抑制平滑肌细胞增殖和促进内皮细胞生长以及血管生成的作用。
OBJECTIVE: To investigate the role of PD-ECGF gene in the prevention of vascular restenosis by transfection of PD-ECGF gene in rabbit carotid artery balloon injury model. Methods Thirty-six New Zealand white rabbits (body weight 2.5-3.0 kg) were used to establish the carotid artery balloon injury model in rabbits. The experimental group (n = 18) and the control group (n = 18) Transfection; control group using delivery agent treatment. Three weeks later, the endothelial coverage and neointima were examined by Evans blue staining and HE staining. The expression of PD-ECGF protein in carotid artery was detected by immunohistochemistry and Western blot. Results The coverage of endothelial cells in the experimental group [(72.8 ± 4.0)%] was significantly higher than that in the control group [(33.6 ± 7.4)%]. HE staining showed that the degree of hyperplasia of intima and media in the experimental group was significantly reduced compared with the control group Western blotting results showed that the expression of PD-ECGF protein in carotid artery of experimental group was significantly higher than that of control group (P <0.05). Conclusion In the rabbit model of carotid artery balloon injury, up-regulation of PD-ECGF gene can inhibit smooth muscle cell proliferation and promote endothelial cell growth and angiogenesis.
