目的:研究γ射线照射后小鼠骨髓造血细胞凋亡及其与 Fas 表达的关系。方法:采用电镜、 D N A 琼脂糖凝胶电泳及流式细胞术观察小鼠经60 Co γ射线照射后不同时间的骨髓造血细胞凋亡及 Fas 的表达情况。结果:在照射后4 h 细胞即出现核固缩、染色质凝集; D N A 琼脂糖凝胶电泳有大量的小分子片段,呈典型“梯状”图谱;流式细胞术分析表明,凋亡细胞百分率在照射后4 h 明显增加,8 h 达到高峰;照后4 h, Fas 表达显著增强。结论:γ射线照射后可引起骨髓造血细胞凋亡, Fas 可能参与了辐射诱导的造血细胞凋亡过程。
OBJECTIVE: To study the apoptosis of bone marrow hematopoietic cells after γ-ray irradiation and its relationship with Fas expression. Methods: The apoptosis of bone marrow hematopoietic cells and the expression of Fas were observed by electron microscopy, D-A agarose gel electrophoresis and flow cytometry after 60 Co γ -ray irradiation. Results: After 4 h of irradiation, the cells showed nuclear condensation and chromatin condensation. There were a large number of small molecular fragments on D N A agarose gel electrophoresis, showing a typical “ladder” pattern. Flow cytometry analysis showed that apoptosis The percentage of cells increased significantly at 4 h after irradiation and peaked at 8 h, and the expression of Fas at 4 h after irradiation significantly increased. Conclusion: γ-ray irradiation can cause apoptosis of hematopoietic cells in bone marrow, and Fas may be involved in the radiation-induced apoptosis of hematopoietic cells.
