目的为了解健康成人和放射工作者体细胞HPRT基因位点突变率。方法应用人外周血淋巴细胞抗6-TG分析技术检测了30例健康成人和30例放射工作者体细胞HPRT位点突变频率。结果健康成人HPRT基因位点突变频率均值为01628×10-3,变化范围为01300×10-4~02903×10-3,放射工作HPRT基因位点的突变频率均值为02225×10-3,变化范围为0337×10-4~0478×10-3。t检验P<005,两者差异显著。从检测结果亦可看出,随着放射工龄的延长,HPRT基因位点突变频率也随之增加。结论体细胞HPRT基因位点突变频率可作为各种有害因素对机体遗传损伤的生物标识物之一。
Objective To understand the mutation rate of somatic HPRT gene loci in healthy adults and radiologists. Methods Human peripheral blood lymphocyte anti-6-TG assay was used to detect the frequency of somatic HPRT mutation in 30 healthy adults and 30 radiologists. Results The average frequency of HPRT gene mutation in healthy adults was 01628 × 10-3, with a range of 01300 × 10-4 ~ 02903 × 10-3. The average frequency of mutations in HPRT gene loci was 0 2225 × 10-3, the variation range is 0337 × 10-4 ~ 0478 × 10-3. t test P <0 05, significant difference between the two. From the test results can also be seen, with the extension of the length of radiation, HPRT gene mutation frequency also increases. Conclusion The frequency of somatic HPRT gene mutation can be used as one of the biomarkers for genetic damage caused by various harmful factors.
