目的探讨超临界CO2流体萃取白花蛇舌草中三萜类成分的可行性。方法以齐墩果酸量、总三萜量及固形物量为考察指标,采用正交试验优化超临界CO2流体萃取白花蛇舌草三萜类成分的工艺条件。同时采用MTT法测定不同提取工艺的白花蛇舌草提取物对A549细胞增殖的影响。结果超临界CO2流体萃取白花蛇舌草三萜类成分的最佳工艺条件为萃取压力15 MPa,萃取温度40℃,夹带剂(95%乙醇)用量为2.0 mL/g,萃取时间1.5 h。超临界萃取物能有效抑制A549细胞的增殖。结论超临界CO2流体可有效萃取白花蛇舌草中三萜类成分。
Objective To investigate the feasibility of extracting triterpenoids from Hedyotis diffusa Willd by supercritical CO2 fluid. Methods Taking oleanolic acid, total triterpene and solid content as indexes, the orthogonal experiment was used to optimize the technological conditions for extracting triterpenoids from Hedyotis diffusa with supercritical CO2 fluid. At the same time, MTT assay was used to determine the effects of different extracts of Hedyotic diffusa on A549 cell proliferation. Results The optimal extraction conditions for triterpenoids from Hedyotis diffusa will be 15 MPa, extraction temperature 40 ℃, entrainer (95% ethanol) 2.0 mL / g and extraction time 1.5 h. Supercritical fluid extract can effectively inhibit the proliferation of A549 cells. Conclusion Supercritical CO2 fluid can effectively extract triterpenoids from Hedyotis diffusa.
