目的:观察川东獐牙菜(口山)酮对大鼠缺血再灌注损伤心肌的保护作用。方法:离体大鼠心脏全心停灌缺血20min后再灌注40min和在体大鼠通过结扎左冠状动脉前降支1h后再灌注3h产生缺血再灌注损伤。离体心脏观测心功能(左室内压、左室内压变化最大速率±dp/dt_(max)、心率)、冠脉流量,测定肌酸激酶(CK)释放量和心肌组织丙二醛(MDA)含量;在体大鼠观测心肌梗死面积和CK释放量。结果:(口山)酮(90和300μg/L)能显著促进再灌注时心功能恢复,减少CK释放和降低MDA的含量。(口山)酮(0.5和1.0mg/kg)均可显著缩小心肌梗死面积和降低CK浓度。结论:川东獐牙菜(口山)酮具有抗心肌缺血再灌注损伤的作用,其保护作用可能与抗脂质过氧化有关。
OBJECTIVE: To observe the protective effect of Scutellaria chuanxiong (Keishan) ketone on myocardial ischemia-reperfusion injury in rats. METHODS: Ischemia-reperfusion injury was induced in isolated rat hearts by cardiac arrest after 20 min of ischemia followed by reperfusion for 40 min and in vivo by ligation of the left anterior descending coronary artery for 1 h followed by 3 h of reperfusion. Heart function (left ventricular pressure, maximum rate of left ventricular pressure change ±dp/dt_(max), heart rate), coronary flow, cardiac creatine kinase (CK) release, and myocardial tissue malondialdehyde (MDA) were measured in isolated hearts. Content; in vivo myocardial infarction area and CK release were observed in rats. RESULTS: (Koushan) ketone (90 and 300 μg/L) significantly promoted cardiac function recovery, reduced CK release and decreased MDA content during reperfusion. (Koushan) Ketone (0.5 and 1.0 mg/kg) both significantly reduced myocardial infarct size and decreased CK concentration. Conclusion: Chuandong Scutellariae (Koushan) Ketone has anti-myocardial ischemia reperfusion injury, and its protective effect may be related to anti-lipid peroxidation.
