β-葡萄糖苷酶(β-glucosidase,BG)是昆虫降解纤维素起关键作用的限速酶,在糖原降解和糖类代谢方面具有重要的生理功能。本研究用粗纤维含量不同的饲料饲养家蝇Musca domestica,分为对照组(粗纤维含量为30%)、实验组1(粗纤维含量为40%)和实验组2(粗纤维含量为50%)。分别在家蝇发育的不同时期取样,采用实时荧光定量PCR技术和DNSA法检测β-葡萄糖苷酶基因的mRNA表达水平和β-葡萄糖苷酶活性。结果显示,β-葡萄糖苷酶基因mRNA表达水平和酶活性随家蝇的个体发育逐渐提高(P<0.05),并且与饲料中粗纤维的含量呈正相关(P<0.05)。实验组1β-葡萄糖苷酶基因mRNA表达水平较对照组提高了1.7~2.4倍,酶活性提高了1.04~1.37倍;实验组2mRNA表达水平较对照组提高了3.1~5.0倍,酶活性提高了1.19~2.25倍。这些结果表明取食粗纤维对家蝇β-葡萄糖苷酶基因表达量及酶活性具有诱导作用。
β-glucosidase (BG) is a rate-limiting enzyme that plays an important role in the degradation of cellulose by insects. It has important physiological functions in glycogen degradation and carbohydrate metabolism. Musca domestica was divided into control group (crude fiber content of 30%), experimental group 1 (crude fiber content of 40%) and experimental group 2 (crude fiber content of 50% ). The samples were collected from different periods of housefly development. The mRNA expression and β-glucosidase activity of β-glucosidase gene were detected by real-time fluorescence quantitative PCR and DNSA. The results showed that the mRNA and protein levels of β-glucosidase increased with the individual development of housefly (P <0.05), and positively correlated with the content of crude fiber in feed (P <0.05). Compared with the control group, the mRNA expression of 1β-glucosidase increased by 1.7-2.4 folds and the activity increased by 1.04-1.37 times. The expression of 2 mRNA in the experimental group increased by 3.1-5.0 folds compared with the control group, and the activity of the enzyme increased by 1.19 ~ 2.25 times. These results indicate that crude fiber feeding induces β-glucosidase gene expression and enzyme activity in housefly.
