[目的]优化南极适冷菌Rheinheimera sp.97产抗菌活性物质的发酵条件。[方法]通过单因素和正交试验对南极适冷菌R.sp.97产抗菌活性物质的发酵培养基进行了优化,并通过单因素试验对其发酵条件进行了优化。[结果]菌株R.sp.97产抗菌活性物质的最佳培养基为:蛋白胨3.0g/L,硫酸铵1.0g/L,淀粉2.0g/L,NaCl15.0g/L;最佳发酵条件为:发酵培养基初始pH8,发酵温度10℃,摇瓶装液量30.0%(V/V),接种量1.0%(V/V)。优化后菌株R.sp.97发酵液的抗菌活性较优化前提高了18.1%。[结论]为研究南极适冷菌R.sp.97的活性物质奠定了基础。
[Objective] The research aimed to optimize the fermentation conditions of Rheinheimera sp.97 antibacterial active substance. [Method] The fermentation medium of R. Antarctic bacteria R.sp. 97 producing antibacterial active substance was optimized by single factor and orthogonal experiment. The fermentation conditions were optimized by single factor experiment. [Result] The optimum culture medium for producing antibacterial active substance of strain R.sp.97 was: peptone 3.0g / L, ammonium sulfate 1.0g / L, starch 2.0g / L, NaCl15.0g / L. The optimum fermentation conditions were : Initial pH8 fermentation medium, fermentation temperature 10 ℃, shake flask liquid volume 30.0% (V / V), inoculum size 1.0% (V / V). The antibacterial activity of the optimized strain R.sp.97 was 18.1% higher than that before optimization. [Conclusion] The study laid the foundation for the study on the active substance of R.sp.97 of Antarctic cold adapted bacteria.
