目的:探讨野百合碱(MCT)腹腔注射建立新西兰兔慢性肺动脉高压(CPH)模型的可行性。方法:将84只新西兰兔随机分为两组,即对照组(24只)和模型组(60只)。在相同的饲料和饲养条件下,模型组腹腔注射MCT溶液60 mg/kg,对照组注射相同剂量的乙醇与生理盐水混合液,28 d后进行右心导管压力测定肺动脉的压力。结果:右心导管各心腔压力的测定值显示,模型组肺的动脉收缩压、舒张压,右室收缩压、舒张压及右房收缩压、舒张压与对照组比较均呈升高的趋势,但仅右房舒张压的差异具有统计学意义(P<0.01);模型组的平均肺动脉收缩压未达到30 mmHg,说明模型的制备不理想。病理检测的结果显示,与对照组比较,模型组兔肺小动脉血管的管壁增厚,平滑肌组织增殖明显。结论:MCT注射可致兔肺动脉血管平滑肌层增厚,造成血流动力学改变,但所致肺动脉压力升高的结果并不稳定,仍需进一步研究。
Objective: To investigate the feasibility of establishing a model of chronic pulmonary hypertension (CPH) in New Zealand rabbits by intraperitoneal injection of monocrotaline (MCT). Methods: 84 New Zealand white rabbits were randomly divided into two groups: control group (24) and model group (60). Under the same feed and feeding conditions, the model group was intraperitoneally injected with MCT solution 60 mg / kg. The control group was injected with the same dose of ethanol and saline solution. The pressure of the pulmonary artery was measured by right heart catheterization 28 days later. Results: The measured values of right ventricular cardiac pressure showed that pulmonary artery systolic pressure, diastolic pressure, right ventricular systolic pressure, diastolic pressure and right atrial systolic pressure, diastolic pressure of the model group were all increased compared with the control group , But only the right atrium diastolic pressure difference was statistically significant (P <0.01); the model group average pulmonary artery systolic pressure did not reach 30 mmHg, indicating that the model preparation is not ideal. The results of pathological examination showed that compared with the control group, the walls of the pulmonary arterioles in the model group were thickened and the proliferation of smooth muscle tissue was obvious. CONCLUSION: MCT injection can cause thickened rabbit pulmonary arterial smooth muscle layer and result in hemodynamic changes. However, the results of pulmonary hypertension caused by MCT injection are not stable and need further study.
