目的了解外周血中白细胞介素8(interleukin-8,IL-8)的表达水平在丙型肝炎病毒(hepatitis C virus,HCV)各感染状态中的差异。方法收集了174例静脉吸毒人群的血样,根据抗-HCV抗体、HCV RNA的检测结果,将HCV感染组分为急性感染组(HCV Ig G-RNA+),慢性感染组(HCV Ig G+RNA+)和自限清除组(HCV Ig G+RNA-),并根据HBs Ag的检测结果将其分为HCV感染组、HBV感染组、HCVHBV共感染组和对照组,并采用悬液芯片检测其外周血中IL-8水平,分析HCV各感染状态的IL-8水平的差异,及在HCVHBV共感染中,HBV对HCV IL-8表达水平影响。结果在HCV各感染组中,急性感染组(HCV Ig G-RNA+)和自限清除组(HCV Ig G+RNA-)的IL-8水平显著高于慢性感染组(HCV Ig G+RNA+)组与对照组(P<0.05);HCV单感染组和HCV HBV共感染中的IL-8水平无显著性差异(P>0.05);IL-8水平与ALT、AST无相关性,IL-8水平在两组不同的HCV基因型人群(3型与6型)中的表达水平相似,无显著差异(P>0.05)。结论外周血中IL-8可能对HCV病毒清除有一定作用,在HCV感染的早期IL-8可能通过参与天然免疫抗病毒,在恢复状态下可能与记忆细胞免疫反应有关,并且HCV感染时IL-8的表达水平不受HBV感染的影响。
Objective To investigate the difference of the expression of interleukin-8 (IL-8) in peripheral blood among different infection states of hepatitis C virus (HCV). Methods The blood samples from 174 intravenous drug users were collected. According to the results of anti-HCV antibodies and HCV RNA, the HCV infection was divided into HCV Ig G-RNA +, HCV Ig G + RNA + (HCV Ig G + RNA-) group were divided into HCV infection group, HBV infection group, HCVHBV co-infection group and control group according to the detection results of HBsAg. The peripheral blood was detected by suspension chip IL-8 levels in HCV infection, and analyze the difference of IL-8 levels among HCV infection states and the effect of HBV on HCV IL-8 expression in HCVHBV co-infection. Results HCV each infection, the acute infection (HCV Ig G-RNA +) and self-limiting clear group (HCV Ig G + RNA-) of IL-8 levels were significantly higher than in chronic infection (HCV Ig G + RNA +) group with the control group (P <0.05); no HCV single infection and HCV HBV IL-8 levels of co-infection of a significant difference (P> 0.05); IL-8 levels of ALT, AST no correlation, IL-8 levels The expression levels in two different HCV genotypes (type 3 and type 6) were similar, with no significant difference (P> 0.05). Conclusion IL-8 in peripheral blood may play an important role in HCV clearance. In the early stage of HCV infection, IL-8 may be involved in innate immune antiviral activity and may be related to memory cell immune response in recovery state. Moreover, IL- 8 expression levels are not affected by HBV infection.
