目的观察CD2相关蛋白(CD2AP)mRNA在大鼠阿霉素肾病模型中的表达,探讨其在蛋白尿发生发展过程中的作用。方法建立大鼠阿霉素肾病模型,应用光镜、电镜观察不同时期(第2、4、6、8周末)肾组织的病理改变,并采用半定量RTPCR的方法,检测大鼠肾皮质中CD2APmRNA的表达。结果阿霉素注射后:(1)大鼠24h尿蛋白量逐渐增加,第2、4、6、8周末分别为(144.5±16.4)、(250.0±45.5)、(440.1±62.1)和(626.5±94.2)mg/24h,均显著高于正常对照组(24.3±6.5)mg/24h(P<0.001);(2)大鼠肾皮质CD2APmRNA的表达在第4、6、8周末分别达到正常对照组的1.7倍、2.5倍和1.9倍。CD2APmRNA的表达与尿蛋白量呈正相关(r=0.58,P<0.01)。结论阿霉素肾病模型大鼠肾皮质CD2APmRNA的表达与大鼠蛋白尿的发生发展具有一定的相关性。
Objective To investigate the expression of CD2AP mRNA in rat adriamycin nephropathy model and its role in the development of proteinuria. Methods The rat adriamycin nephropathy model was established. The pathological changes of renal tissues in different periods (2nd, 4th, 6th and 8th week) were observed by light microscope and electron microscope. The semi-quantitative RTPCR method was used to detect the expression of CD2AP mRNA expression. Results After injection of doxorubicin, the amount of urinary protein in 24h increased gradually (144.5 ± 16.4), (250.0 ± 45.5), (440.1 ± 62.1) and (626.5 ± 94.2) mg / 24h, all of which were significantly higher than that of the normal control group (24.3 ± 6.5) mg / 24h (P <0.001). (2) The expression of CD2AP mRNA in renal cortex reached the normal control 1.7 times, 2.5 times and 1.9 times of the group. CD2AP mRNA expression was positively correlated with urine protein (r = 0.58, P <0.01). Conclusion The expression of CD2AP mRNA in renal cortex of rats with adriamycin nephropathy may be related to the occurrence and development of proteinuria in rats.
