目的 :研究大鼠脑缺血再灌注后生长相关蛋白 - 4 3( GAP- 4 3)表达的变化规律及外源性神经生长因子 ( NGF)的影响。方法 :成年健康雌性 Wistar大鼠 72只 ,随机分为假手术组、自然恢复组、人工脑脊液组和 NGF治疗组。采用线栓法建立大脑中动脉缺血再灌注 ( MCAO)动物模型 ,应用免疫组织化学方法观察脑缺血再灌注后 GAP- 4 3的表达。结果 :( 1)脑缺血再灌注 6h后 ,缺血周围区 GAP- 4 3表达逐渐增高 ,第 7天达高峰 ,以后逐渐降低 ,第 2 1天仍有表达。 ( 2 )应用外源性 NGF后 ,GAP- 4 3表达较对照组有所增高 ,但无显著性差异 ( P>0 .0 5)。结论 :提示中枢神经系统损伤后 ,神经元具有再生和修复的可塑性 ,外源性 NGF对 GAP- 4 3的调节有待于进一步研究
Objective: To study the change of expression of GAP-43 and the effect of exogenous nerve growth factor (NGF) after cerebral ischemia-reperfusion in rats. Methods: Seventy-two healthy female Wistar rats were randomly divided into sham operation group, natural recovery group, artificial cerebrospinal fluid group and NGF treatment group. The animal model of middle cerebral artery occlusion (MCAO) was established by thread occlusion. The expression of GAP-43 after cerebral ischemia-reperfusion was observed by immunohistochemistry. Results: (1) After 6h of cerebral ischemia and reperfusion, the expression of GAP-43 in ischemic area gradually increased and peaked on the 7th day, then gradually decreased and remained on the 21st day. (2) The expression of GAP-4 3 increased with the application of exogenous NGF, but there was no significant difference (P> 0.05). CONCLUSION: Neurons have the plasticity of regeneration and repair after CNS injury, and the regulation of GAP-43 by exogenous NGF remains to be further studied
