目的观察结缔组织生长因子(CTGF)反义寡核苷酸对TGF-β1诱导的肾小管上皮细胞NRK52E胶原Ⅲ分泌的影响。方法采用脂质体介导的方法将CTGF反义寡核苷酸导入细胞,转化生长因子β1(TGF-β1)刺激48 h后,用RT-PCR、ELISA法观察CTGF和胶原Ⅲ的表达。结果TGF-β1能够诱导NRK52E细胞表达CTGF并进而促进胶原Ⅲ的分泌,CTGF反义寡核苷酸导入细胞后可以大部分取消TGF-β1的作用,而对照组的寡核苷酸没有此作用。结论CTGF反义寡核苷酸能够特异地抑制CTGF的表达,进而阻止细胞外基质的生成,这可能是治疗肾间质纤维化的一条新途径。
Objective To observe the effects of connective tissue growth factor (CTGF) antisense oligonucleotide on the secretion of collagen Ⅲ of NRK52E induced by TGF-β1 in renal tubular epithelial cells. Methods CTGF antisense oligonucleotides were introduced into cells by liposome-mediated method. The expression of CTGF and collagen Ⅲ was detected by RT-PCR and ELISA after stimulated by transforming growth factor-β1 (TGF-β1) for 48 h. Results TGF-β1 induced the expression of CTGF in NRK52E cells and promoted the secretion of collagen Ⅲ. However, most of the effects of TGF-β1 were abolished after CTGF antisense oligonucleotides were introduced into the cells, while the control oligonucleotides did not. Conclusion CTGF antisense oligonucleotide can specifically inhibit the expression of CTGF, thereby preventing the extracellular matrix formation, which may be a new way to treat renal interstitial fibrosis.
