目的观察清毒汤对实验性重症肝损伤大鼠丙氨酸氨基转移酶(alanine aminotransferase,ALT)、内毒素、凝血酶原活动度(prothrombin activity,PTA)及D-乳酸表达的影响,探讨该方治疗慢性重型肝炎内毒素血症的作用机制。方法建立TAA致实验性大鼠重症肝损伤模型,清毒汤干预后,检测大鼠ALT、内毒素、PTA及D-乳酸的表达水平。结果与正常组比较,模型组大鼠ALT、内毒素、D-乳酸水平均显著升高(P<0.01),PTA水平显著下降(P<0.01);与模型组比较,清毒汤低、中剂量组及乳果糖组血清ALT、内毒素、PTA及D-乳酸表达水平均有不同程度的改善(P<0.05),清毒汤高剂量组除血清内毒素含量外各指标均明显改善(P<0.05);与乳果糖组比较,清毒汤中剂量组除PTA外各指标均不同程度降低(P<0.05),低剂量组各指标均无明显差异(P>0.05),高剂量组除PTA外各指标明显高于乳果糖组(P<0.05)。结论清毒汤对实验性肝损伤大鼠有较好的改善肝损伤和降低内毒素表达的作用,尤其中剂量组最为明显,推测其作用机制可能与降低肠道通透性相关。
Objective To observe the effect of Qingdu Decoction on the expression of alanine aminotransferase (ALT), endotoxin, prothrombin activity (PTA) and D-lactate in rats with experimental severe liver injury Fang treatment of chronic severe hepatitis endotoxemia mechanism of action. Methods To establish a rat model of severe liver injury induced by TAA. After intervention of Qingdu Tang, the levels of ALT, endotoxin, PTA and D-lactate in rats were determined. Results Compared with the normal group, the levels of ALT, endotoxin and D-lactate in the model group were significantly increased (P <0.01), and the PTA level was significantly decreased (P <0.01). Compared with the model group, The levels of serum ALT, endotoxin, PTA and D-lactate in the dosage group and lactulose group were all improved to some extent (P <0.05), and the indexes in the high-dose Qingqintang group except serum endotoxin were significantly improved (P (P <0.05). Compared with lactulose group, each index of Qingdu Tang medium dose group except PTA decreased to some extent (P <0.05), but there was no significant difference between low dose group and high dose group Each index outside PTA was significantly higher than that of lactulose group (P <0.05). Conclusions Qingdu decoction can improve liver injury and reduce endotoxin expression in experimental liver injury rats, especially in the middle dose group, suggesting that its mechanism of action may be related to reducing intestinal permeability.
